Summary: The expression of KLHL21 is significantly reduced in bladder carcinoma tissues and cell lines, and lower expression of KLHL21 is associated with poorer overall survival. Over-expression of KLHL21 inhibits the proliferation, migration and invasion of bladder carcinoma cells, while knockdown of KLHL21 accelerates these processes. KLHL21 negatively regulates the activation of the NF-KB signaling pathway and down-stream molecules involved in cell survival, death, and migration.

Summary: NF-KB signaling is active in pancreatic cancer and plays a crucial role in its progression. This study demonstrates that VRK2 regulates TNFa/NF-KB signaling by interacting with IKKss and activating the pathway. VRK2 overexpression promotes the growth of pancreatic cancer cells and organoids. Inhibition of IKKss abolishes the promotive effect of VRK2. These findings suggest that VRK2 may serve as a potential therapeutic target for pancreatic cancer.

Summary: Muscle-invasive bladder cancer (MIBC) exhibits strong inter- and intra-tumor heterogeneity and mutations in RTK-RAS-PI3K and P19-P53-P21 pathways are commonly observed. This study shows that deficiencies of both PTEN and P53 are responsible for driving the basal/squamous subtype MIBC. PTEN is inactivated by hyperphosphorylation of the C-terminus and this modification may serve as a biomarker for subtyping MIBC and predicting tumor progression. Tailless PTEN is a potential molecular therapeutic for accessible tumors like bladder cancer.

Summary: Noncoding RNAs play a significant role in human cancer development, but their involvement in bladder cancer is not well understood. This study revealed that the long noncoding RNA lnc-STYK1-2 inhibits bladder cancer cell proliferation, migration, and tumorigenesis by targeting miR-146b-5p to regulate ITGA2 expression and AKT/STAT3/NF-kB signaling pathways. Additionally, the study demonstrated that lnc-STYK1-2 can directly associate with miR-146b-5p, providing insight into the molecular mechanisms underlying bladder cancer progression.

Summary: This study explored the functional mechanism of action of LINC00460 in cervical cancer angiogenesis for the first time. The results showed that conditioned medium from LINC00460-knockdown cervical cancer cells attenuated human umbilical vein endothelial cell (HUVEC) migration, invasion, and tube formation, while LINC00460 upregulation had the opposite effects. Mechanistically, LINC00460 stimulated VEGFA transcription. The study suggests that the NF-KB-VEGFA axis activated by LINC00460 is a promising target for blocking tumor angiogenesis.
