Summary: Human RNase MRP ribonucleoprotein complex is an essential endoribonuclease involved in processing various types of RNA. Recent structural studies have shed light on the catalytic mechanism of yeast RNase MRP and related RNase P, but the mechanism of RMRP in human RNase MRP complex remains unclear. The crystal structure reveals the binding of RMRP's P3 domain to the RPP20-RPP25 proteins and shows a potential role in mediating the dimerization of the complex. This structure also provides insights into the assembly and pathogenesis of human RNase MRP complex.

Summary: The cartilage hair hypoplasia and anauxetic dysplasia (CHH-AD) spectrum includes rare skeletal disorders, with anauxetic dysplasia (ANXD) being the most severe. Biallelic variants in RMRP, POP1, and NEPRO have been associated with ANXD types. This study describes a new ANXD3 patient with unique clinical features, expanding the phenotypic spectrum of ANXD. Greater awareness of these features may improve early diagnosis and treatment.

Summary: Eugenol, an essential oil extracted from cloves, has analgesic and anesthetic effects and is commonly used in fishery anesthesia. However, the extensive use of eugenol in aquaculture production may pose safety risks and developmental toxicity during the early life stages of fish. This study found that eugenol exposure delayed the hatching of zebrafish embryos, decreased body length and inflation rate of the swim bladder, and increased the mortality rate of zebrafish larvae. Furthermore, eugenol inhibited the Wnt/beta-catenin signaling pathway involved in swim bladder development, leading to abnormal development and impaired feeding ability in zebrafish larvae.

Summary: This study evaluated the immunotoxic and behavioral effects of sanguinarine exposure on larval zebrafish. The results showed that exposure to sanguinarine led to shorter body length, larger yolk sacs, slower heart rates, reduced number of immune cells, and altered locomotor behavior. In addition, it caused oxidative stress and increased apoptosis, as well as aberrant expression of key genes in the TLR immune signaling pathway. Overall, sanguinarine exposure may cause immunotoxicity and aberrant behavior in larval zebrafish.

Through analysis of nine childhood-onset patients with biochemically and genetically unknown PAI, various-sized single-exon deletions involving ZNRF3 exon 2 were identified. Further research showed that these deletions could lead to the inactivation of RSPO1-dependent Wnt/β-catenin signaling.

Summary: The neural crest (NC) is a multipotent cell population in vertebrate embryos with extraordinary migratory capacity, crucial for vertebrate development. The ongoing role of the Wnt/beta-catenin signalling pathway during NC development, especially in the induction of NC and specification of melanocytes, is discussed. Current understanding shows the continual involvement of Wnt/beta-catenin signalling in activating and maintaining the gene regulatory network during NC induction and pigment cell specification, with implications for emerging models and hypotheses on NC fate restriction.
