The maximal attained yeast biomass concentration was close to 70 g/L, with a 2-PE biosynthesis rate of 0.372 g/L/h and a yield of 74%.

Summary: This study developed a strategy to maximize the biomass concentration and lipid accumulation by Nigrospora sp. in submerged fermentation. Through optimizing the media compositions and process variables, the maximum biomass concentration and lipid accumulations reached 40.17 g/L and 21.32 wt% in the bioreactor, which were 2.1 and 5.4 times higher than those in shaken flasks, respectively.

Summary: Process intensification is gaining popularity in the mammalian biomanufacturing industry as a result of the increasing demand for biotherapeutics. This study established a CHO cell process and investigated the impact of butyric acid supplementation on the production process. The findings showed that butyric acid supplementation led to higher cellular productivities and had minimal effects on product quality attributes. Additionally, detailed transcriptomic analysis revealed potential mechanisms for gene engineering optimization.

Summary: This study investigated the effects of four feeding medium factors on the culture efficiency of Oplopanax elatus adventitious roots, and identified the optimal conditions for sucrose concentration, salt strength, phosphorus, and nitrogen concentration. The findings provide a reference for further improving the fed-batch culture system of O. elatus adventitious roots. The application of the analytic hierarchy process method lays a foundation for scientifically and reliably analyzing data with multiple indexes in the field of plant cell culture.

Summary: This study focuses on the fermentative production of mannosylerythritol lipids (MEL) in a bioreactor with Moesziomyces aphidis. By using an exponential fed-batch strategy, the biomass concentration was increased, leading to higher MEL formation rates. The maximum MEL concentration obtained was 50.5 g/L when excess oil was added, but with high concentrations of residual fatty acids present in the broth.

Summary: Continuous cultivation of mammalian cells has evolved into high-density perfusion processes, achieving volumetric productivities over 5 g L-1 day(-1). Advancements in downstream processing and automation have led to the integration of drug substance manufacturing processes suitable for next-generation plants. Perfusion has also been used for high-density cell bank generation and seed-train optimization, resulting in significant productivity increases.

Summary: In this study, the mechanical properties of Escherichia coli (E. coli) cells with different physiological states were investigated using atomic force microscopy. It was found that the peptide-producing cells were softer and had a lower Young's modulus compared to the non-producing cells, and this difference increased over time. This study provides the first evidence that the metabolic burden and carbon limitation significantly influence the physical properties (mechanical properties) of cells in bioreactors.

Summary: This study achieved high-yield hydrolysis of corncob with 25% solids content using different pretreatment methods and fed-batch modes. The optimal feeding mode was found to be a low amount and high frequency mode, resulting in 84.4% glucose yield in 96 hours. This research demonstrates that the combination of optimized pretreatment method and fed-batch mode is favored for industrial-scale high-solids hydrolysis.
