Linifanib exerts dual anti-obesity effect by regulating adipocyte browning and formation

Obesity is caused by energy imbalance and accompanied by adipocyte hypertrophy and hyperplasia. Therefore, both enhancement of adipocyte energy expenditure and inhibition of adipogenesis are viable ways to combat obesity. Using the Ucp1-2A-luciferase reporter animal model previously reported by us as a screening platform, a chemical compound Linifanib was identified as a potent inducer of UCP1 expression in primary inguinal adipocytes in vitro and in vivo. Signal pathway analyses showed that Linifanib promoted adipocyte browning by attenuating STAT3 phosphorylation. The effects of Linifanib on adipocyte browning were blocked by the compound, SD19, which activates the STAT3 signaling cascade. Linifanib also inhibited adipocyte differentiation, by blocking mitotic clonal expansion, which could be rescued by STAT3 activator. Taken together, our results indicate that Linifanib might serve as a potential drug for the treatment of obesity.

The shades of grey in adipose tissue reprogramming

Summary: Adipose tissue plays a crucial role in obesity-related pathologies through regulating systemic immunometabolism. It exhibits remarkable plasticity during obesity, affecting tissue morphology, extracellular matrix composition, angiogenesis, immunometabolic balance, and circadian rhythmicity. Dysregulation in these features escalates the pathogenesis caused by adipose tissue. The potential to be reprogrammed appears to be a target for the treatment of obesity and its associated comorbidities.

Transforming primary human hepatocytes into hepatocellular carcinoma with genetically defined factors

Summary: The lack of in vivo models has hindered our understanding of hepatocellular carcinoma (HCC) development. In this study, we used genetic screening in an immunodeficient mouse model to identify oncogenes highly expressed in induced HCC (iHCC) samples. Overexpression of these genes transformed primary human hepatocytes into iHCC. The iHCC model recapitulates histological architecture and gene expression characteristics of clinical HCC samples, providing a biological basis for studying HCC initiation and identifying diagnostic biomarkers and targets for cellular immunotherapy.

Generation of a genetically modified pig model with CREBRFR457Q variant

Summary: Obesity is a strong risk factor for type 2 diabetes. This study found that the CREBRF variant is associated with obesity and reduced risk of T2D in people of Pacific ancestry. By introducing the CREBRF variant into the porcine genome, researchers observed that the variant led to significantly increased fat deposition, mainly in subcutaneous adipose tissue. The variant also promoted preadipocyte differentiation, resulting in a greater number but smaller size of adipocytes in subcutaneous adipose tissue. Additionally, the variant pigs showed reduced oxidative stress in adipose tissue, indicating a relative metabolic protection despite their obesity.

Adipocytes release lactate as a signal of inflammation, which can enhance obesity-associated inflammation through direct binding with PHD2. This study reveals the critical role of adipocyte-derived lactate in promoting the pro-inflammatory microenvironment in adipose tissue and identifies PHD2 as a direct sensor of lactate, linking chronic inflammation and energy metabolism.

Androgen Receptor is a Negative Regulator of PRDM16 in Beige Adipocyte

Summary: PRDM16 is a dominant activator of brown and beige adipocytes. The regulation of PRDM16 expression is not fully understood. Androgen receptor (AR) is negatively correlated with PRDM16 expression. Sex differences in PRDM16 mRNA expression are observed in human WAT, with females having higher expression than males. Androgen-AR signaling inhibits PRDM16 expression and beige adipocyte browning. AR directly binds to the intronic region of PRDM16, but not to Ucp1 and other browning-related genes. Adipocyte-specific deletion of Ar promotes beige cell biogenesis, while adipocyte-specific overexpression of AR attenuates white adipose beiging.

Cold-Induced Reprogramming of Subcutaneous White Adipose Tissue Assessed by Single-Cell and Single-Nucleus RNA Sequencing

Summary: Through transcriptomic profiling at the single-cell and single-nucleus resolution, we reconstructed the cellular atlas in mouse subcutaneous white adipose tissue and revealed the transcriptomes and intercellular cross-talks during brown remodeling. Our findings identified subpopulations in mature adipocytes, adipose stem and progenitor cells, and endothelial cells, and uncovered their interconversion and reprogramming in response to cold. Our data unveil fundamental changes during cold-evoked adipose browning.
