Summary: Plasmacytoid dendritic cells (pDC) are the main type I interferon-producing cells in humans and are capable of regulating innate and adaptive immune responses. Tumor infiltration by pDC is well-documented in various cancers and is associated with unfavorable clinical outcomes. While Blastic Plasmacytoid Dendritic Cells Neoplasm (BPDCN) is a well-described aggressive leukemia derived from pDCs, the understanding of tumor infiltration by mature pDCs in Myeloid Neoplasms (MN) is limited. This review aims to provide a comprehensive overview of pDC biology in MN, emphasizing the need for further research into pDC oncogenesis and immune responses for better therapeutic strategies.

Recent studies have challenged the traditional view of neutrophils as static responders with a short lifespan, revealing their heterogeneity and dynamic response. This article focuses on discussing their involvement in aging and neurological disorders, particularly emphasizing their impact on chronic inflammation and contribution to neurological diseases. It concludes that reactive neutrophils directly contribute to increased vascular inflammation and age-related diseases.

Summary: Failure of bone healing after fracture often results in nonunion, but the underlying mechanism of nonunion pathogenesis is poorly understood. This study reveals that the inflammatory microenvironment in atrophic nonunion inhibits the expression of DNA methyltransferases, resulting in overexpression of CtBP1/2. The increased CtBP1/2 acts as transcriptional corepressors that suppress the expression of six genes involved in bone healing. The disruption of this signaling may represent a new therapeutic strategy to prevent nonunion incidence after bone fracture.
