The synergistic effect of PFK15 with metformin exerts anti-myeloma activity via PFKFB3

High glucose metabolism provides sufficient energy for cancer cells and is enabled by metabolic enzymes. PFKFB3 (6-phosphofructo-2-kinase) accelerates the synthesis of fructose 2,6-bisphosphate (F2,6P(2)), which is a powerful allosteric regulatory activator of 6-phosphofructo-1-kinase (PFK-1), a rate-limiting enzyme of glycolysis. The aim of this study was to investigate the anti-myeloma function and underlying mechanism of suppressing PFKFB3 via PFK15 (1-(4-pyridinyl)-3-(2-quinoliny1)-2-propen-1-one). The role of PFK15 in killing multiple myeloma (MM) cells was evaluated by cytotoxicity and apoptosis assays, flow cytometry and Western blotting. The oral hypoglycemic drug metformin (Met) was found to inhibit PFKFB3 protein expression by gene chip and Western blotting techniques in our study. PFK15 also demonstrated a synergistic effect with metformin to eliminate MM cells. Taken together, our findings indicate that PFK15 inhibits MM cell proliferation through the PFKFB3/MAPKs/STAT signaling pathway. The combination therapy of PFK15 and metformin may be a promising anticancer drug regimen for the treatment of MM. (C) 2019 Elsevier Inc. All rights reserved.

Clinical analysis of patients with primary and secondary extranodal natural killer/T-cell lymphoma of central nervous system

Summary: Extranodal natural killer (NK)/T-cell lymphoma (NKTL) rarely involves the central nervous system (CNS) and has a poor prognosis with no standard treatment. In a study of 414 ENKL patients, 4.59% were diagnosed with CNS involvement. 2 patients had primary CNS (PCNS) NKTL and 17 patients had secondary CNS (SCNS) invasion. 9 patients survived with a median overall survival time of 55 months, and the median survival time after CNS invasion was 17 months. The 5-year cumulative survival probability was 45.7%. Evaluation and prophylaxis treatment for CNS risk is recommended for patients with NK/T-cell lymphoma prognostic index risk group III/IV, and systemic therapy based on methotrexate combined with radiotherapy and intrathecal chemotherapy can be considered.

Towards Friction Stir Remanufacturing of High-Strength Aluminum Components

The design of consumable pins had a significant impact on material flow, forging at the repairing interface, load transfer, and fracture characteristics. Proper design criteria resulted in sound repaired components with improved tensile strength and elongation.

Synthesis of TiO2/g-C3N4 Photocatalyst with Recovered TiO2 from Spent SCR Catalyst for Photodegrading Rhodamine B

Summary: In this study, spent SCR catalyst was used to recover TiO2 and modified with g-C3N4 to prepare the R-TiO2/g-C3N4 photocatalyst. XRD patterns confirmed that R-TiO2 maintained the crystal structure of anatase TiO2. The photodegradation efficiency of R-TiO2/g-C3N4 photocatalyst reached 97.11% for Rhodamine B degradation under visible light, which was 1.9 times higher than that of pure TiO2/g-C3N4 photocatalyst.

Experimental study of deep-burial underground structures subjected to multiple 45° side-top far-field explosions

Extremely Sparse Networks via Binary Augmented Pruning for Fast Image Classification

Summary: In this article, the advantages of extremely sparse networks with binary connections for image classification through software-hardware codesign are investigated. A binary augmented extremely pruning method is proposed to achieve high sparsity with minimal accuracy degradation, and a hardware architecture based on the resulting sparse and binary networks is designed to explore the benefits of extreme sparsity with negligible consumption. Experiments on ImageNet classification and FPGA demonstrate a significant tradeoff between accuracy and efficiency in the proposed software-hardware architecture.

GPENs: Graph Data Learning With Graph Propagation-Embedding Networks

Summary: Compact representation of graph data is a fundamental problem in pattern recognition and machine learning. Graph neural networks (GNNs) have been widely studied for graph-structured data representation and learning. In this article, a new model called graph propagation-embedding networks (GPENs) is proposed, which integrates feature propagation and low-dimensional embedding into a unified network. GPENs have good motivation and can be well-explained, and they guarantee both compactness and efficiency. Experiments demonstrate the effectiveness and benefits of GPENs and M-GPENs.

Combined organic-inorganic fertilization builds higher stability of soil and root microbial networks than exclusive mineral or organic fertilization

Summary: This study investigated the impact of different soil types, plant varieties, and fertilizer regimens on soil and root bacterial communities. A nano-liposomal carrier containing p-coumaric acid for induction of targeted apoptosis on melanoma cells and kinetic modeling

They identified two types of oscillations, sarcomeric oscillations and sarcosynced oscillations, and visualized their behavior during propagating waves. The researchers discovered the presence of sarcomeric defect holes and sarcomeric collision holes, which are important indicators for understanding the oscillation properties of sarcomeres. This finding has important implications for improving our understanding of muscle function and its regulatory mechanisms.

Development of a Cre-recombination-based color-switching reporter system for cell fusion detection

TP53INP2 modulates the malignant progression of colorectal cancer by reducing the inactive form of β-catenin

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

The four subunits of rabbit skeletal muscle lactate dehydrogenase do not exert their catalytic action additively

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

Interaction of gallium, indium, and vanadyl curcumin complexes with hen egg-white lysozyme (HEWL): Mechanistic aspects and evaluation of antiamyloidogenic activity

Summary: This study explored the inhibitory effects of gallium curcumin, indium curcumin, and vanadyl curcumin on the amyloid fibrillation of hen egg white lysozyme, as well as the binding interactions of these metal complexes with the enzyme. The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.

The effects of noise exposure on hippocampal cognition in C57BL/6 mice via transcriptomics
