Participation of NADPH Oxidase-Related Reactive Oxygen Species in Leptin-Promoted Pulmonary Inflammation: Regulation of cPLA2 and COX-2 Expression

Obesity is a worldwide epidemic problem and correlates to varieties of acute or chronic lung diseases such as acute respiratory distress syndrome, chronic obstructive pulmonary disease, and pulmonary fibrosis. An increase of leptin, a kind of adipokine, in lean mice plasma has been found to impair immune responses and facilitate the infection of Klebsiella pneumoniae, resulting in increased pneumonia severity. Also, a higher leptin level is found in exhaled breath condensates of obese or asthmatic subjects, compared to healthy ones, suggesting that leptin is involved in the occurrence or exacerbation of lung injury. In previous studies, we showed that leptin stimulated cytosolic phospholipase A2- (cPLA2) gene expression in lung alveolar type II cells via mitogen-activated protein kinase (MAPK) and nuclear factor-kappa B (NF-B)-activated coactivator p300. Herein, we show that the in vivo application of leptin in the respiratory system upregulated the expression of inflammatory proteins cPLA2 and cyclooxygenase-2 (COX-2) together with leukocyte infiltration. Treatment with an ROS scavenger (N-acetylcysteine, NAC), an NADPH oxidase inhibitor (apocynin), or an activating protein (AP)-1 inhibitor (tanshinone IIA) attenuated leptin-mediated cPLA2/COX-2 expression and leukocyte recruitment in the lung. Leptin increased intracellular oxidative stress in a leptin receptor (OB-R) and NADPH oxidase-dependent manner, leading to the phosphorylation of the AP-1 subunit c-Jun. In summation, leptin increased lung cPLA2/COX-2 expression and leukocyte recruitment via the NADPH oxidase/ROS/AP-1 pathway. Understanding the inflammatory effects of leptin on the pulmonary system provides opportunities to develop strategies against lung injury related to metabolic syndrome or obesity.

The GDF15-based death prediction model could potentially be integrated into artificial intelligence-based precision medicine.

Summary: The extension of the cosmic-ray spectrum beyond 1 petaelectronvolt indicates the existence of PeVatrons, cosmic-ray factories that accelerate particles to PeV energies. Detection of ultrahigh-energy gamma radiation is a key signature of PeVatrons, with ongoing efforts to locate and identify these extreme accelerators. Despite detecting high-energy gamma rays from various sources, the exact origin and identity of the PeVatrons responsible for the ultrahigh-energy gamma rays remain unknown.

A Long-Term Study of Alignment Correction Following Proximal Femoral Varus Osteotomy and Pemberton Osteotomy in Children With Legg-Calve-Perthes Disease and Developmental Dysplasia of the Hip

Summary: This study investigated the effect of proximal femoral varus osteotomy (PFVO) on angular deformities in the knees and ankles. The results showed that in patients with Legg-Calve-Perthes disease (LCPD), the femoral neck-shaft angle (FNSA) significantly improved after the procedure, while in patients with developmental dysplasia of the hip (DDH), there was no significant change. Additionally, there were no significant differences in tibiotalar angle (TTA), mechanical lateral distal femoral angle (mLDFA), mechanical axis deviation (MAD), and leg length discrepancy between the operative and non-operative limbs after a 5-year follow-up. This study suggests that PFVO can narrow the gap of angular growth in the knees and ankles between the operative and non-operative limbs in LCPD or DDH patients.

Summary: Uremic vascular calcification (UVC) is closely related to oxidative injury, cell death, and osteocytic conversion. Scavenging ROS may attenuate inflammasome activation and eicosanoid inflammation in the UVC process. Therefore, reducing ROS and PCS could be an effective therapeutic strategy for UVC-related cardiovascular events.

Summary: This study focuses on the characterization of beam ion loss during neutral beam injection (NBI) and proposes methods to improve beam ion confinement. The results show that the dominant mechanisms of beam ion loss are shine-through loss, prompt loss, and stochastic ripple loss. The study provides valuable insights for the improvement of beam ion population confinement and the evaluation of the NBI system on EAST and future tokamaks.

Summary: In this study, the kilometer square array (KM2A) of the large high altitude air shower observatory (LHAASO) was used to analyze dark matter by searching for signals at energies above 10 TeV. The results showed no excess of dark matter signals and placed strong constraints on the lifetime of heavy dark matter particles. These findings have important implications for dark matter interpretations of the diffuse astrophysical high-energy neutrino emission.

Synergistic Chemopreventive Effects of a Novel Combined Plant Extract Comprising Gallic Acid and Hesperidin on Colorectal Cancer

Summary: This study aims to investigate the therapeutic mechanism of a novel combination of gallic acid and hesperidin derived from Hakka pomelo tea (HPT) against colorectal cancer cell growth, including cell viability, cell cycle-associated proteins, spheroid formation, and stemness. The results showed that the combined extract had a higher inhibitory effect on CRC cell viability and may serve as a potential chemopreventive agent.

Prophylactic Effects of Purple Shoot Green Tea on Cytokine Immunomodulation through Scavenging Free Radicals and NO in LPS-Stimulated Macrophages

Summary: This study compared the effects of different tea processes on immune modulation and oxidative stress in macrophages. The results showed that purple shoot tea has potential prophylactic benefits for reducing inflammation and regulating cytokine expression. Purple shoot green tea, in particular, exhibited strong antioxidant properties. Tea extracts were able to reduce inflammation and modulate immune cell responses by regulating cytokine expression.
