Increasing O-GlcNAcylation is neuroprotective in young and aged brains after ischemic stroke

Summary: Our study established the neuroprotective role of XBP1s in ischemic stroke, linked to O-GlcNAcylation, and the therapeutic potential of targeting this axis, particularly in the aged brain. We discovered a novel mechanism involving compromised UDP-GlcNAc availability in the aged brain, contributing to impaired O-GlcNAcylation activation after ischemia. Furthermore, we confirmed the therapeutic effects of glucosamine treatment in both young and aged animals in stroke models, supporting the promise of increasing O-GlcNAcylation in stroke therapy.

Summary: This study demonstrates that ENTPD1 expression on T cells within cutaneous squamous cell carcinomas (cSCCs) is increased, leading to elevated levels of extracellular adenosine in tumors compared to normal skin. Furthermore, higher ENTPD1 expression is associated with metastatic cSCCs. The findings suggest that ENTPD1 regulates DNA damage repair through purinergic signaling, contributing to skin carcinogenesis and metastasis.

Safety Profile of Rapamycin Perfluorocarbon Nanoparticles for Preventing Cisplatin-Induced Kidney Injury

Summary: In this study, the potential of rapamycin perfluorocarbon (PFC) nanoparticles to mitigate cisplatin-induced acute kidney injury was demonstrated with a single preventative dose. The pharmacokinetics and biodistribution of rapamycin were improved by the PFC nanoparticle delivery system. The safety of rapamycin PFC nanoparticles was also confirmed in both in vitro and in vivo experiments.

Summary: This study used a redox-active MnP compound to investigate the redox status of ovarian cancer cells. The results showed that chemotherapy-resistant cells were under higher oxidative stress and had increased glycolysis activity and mitochondrial respiration. Altering the redox state could effectively inhibit the growth and metabolic activity of these cells.

Summary: The study investigates the effects of MnTnHex in non-small cell lung cancer (NSCLC) cells. The results show that MnTnHex can significantly reduce cell viability, alter cell cycle distribution, induce cell death, enhance the cytotoxicity of cisplatin, and reduce cell migration. Metabolomics study also reveals an increase in oxidative stress-related compounds in MnTnHex-treated cells.

Summary: In order to solve the problem of the short half-life of biologics, researchers developed an injectable PEG-like conjugate that forms a subcutaneous depot, providing sustained delivery of the drugs. This conjugate showed better pharmacokinetic and pharmacodynamic properties compared to PEG conjugates and branched PEG, and it also remained effective and safe for long-term management of type 2 diabetes without causing an immune response.

Summary: This article reports the preclinical characterization of a slow, tight-binding LpxC inhibitor, LPC-233, with low picomolar affinity. The compound is a rapid bactericidal antibiotic, unaffected by established resistance mechanisms to commercial antibiotics, and displays outstanding activity against a wide range of Gram-negative clinical isolates in vitro. It is orally bioavailable and efficiently eliminates infections caused by susceptible and multidrug-resistant Gram-negative bacterial pathogens in murine soft tissue, sepsis, and urinary tract infection models.
