Summary: This study aimed to investigate the regulatory role of differentially-expressed circular RNAs (circRNAs) in mouse cardiomyocytes during doxorubicin-induced cardiotoxicity. The expression profiles of circRNAs were significantly altered, with circArhgap12 playing a competitive role among endogenous RNAs in regulating cardiotoxicity. Further analysis revealed that circArhgap12 promoted apoptosis and oxidative stress by sponging miR-135a-5p, suggesting a potential therapeutic target in doxorubicin-induced cardiotoxicity.

Summary: In this study, we found that miR-34a-5p was upregulated in the heart after DOX treatment and inhibiting miR-34a-5p reduced autophagy and pyroptosis in DIC. We also found that inhibiting miR-34a-5p suppressed pyroptosis by regulating autophagy and reducing mitochondrial reactive oxygen species. Moreover, Sirtuin3 (Sirt3) was identified as a target gene of miR-34a-5p.

Inhibition of miR-15b-5p alleviated inflammation, mitochondrial stress, and cell death mechanisms. Furthermore, the inhibition of PCSK9 was found to enhance the protective effect of miR-15b-5p through increased SIRT4 levels.

Summary: Endothelial dysfunction is crucial in vascular disease pathogenesis. Long noncoding RNA (lncRNA) and microRNA (miRNA) play significant roles in various cellular processes in vascular endothelial cells (VECs), including cell growth, migration, autophagy, and apoptosis. The present study investigated the role of plasmacytoma variant translocation 1 (PVT1) in regulating autophagy and apoptosis in human umbilical vein endothelial cells (HUVEC). It was found that PVT1 knockdown accelerates apoptosis and suppresses cellular autophagy. PVT1 functions as a competing endogenous RNA (ceRNA) of miR-15b-5p and miR-424-5p, promoting cellular autophagy and down-regulating apoptosis. This study suggests PVT1 as a potential therapeutic target for cardiovascular disease treatment.

Summary: The study aimed to elucidate the role and mechanism of circMMD in tumor treating fields (TTF) treatment of glioblastoma (GBM). The results showed that circMMD expression was elevated in GBM and high expression was associated with poor prognosis. TTF intervention reduced circMMD synthesis, suppressing GBM proliferation and increasing TTF-mediated apoptosis. The reduction of circMMD promoted the interaction between FUBP1 and FIR, leading to decreased DVL1 transcription. Additionally, decreased circMMD enhanced miR-15b-5p activity to degrade FZD6. Ultimately, the diminished expression of DVL1 and FZD6 suppressed the activation of Wnt/beta-catenin pathway.
