Telomerase is thought to play an essential role in tumorigenesis and progression. Its activity is directly correlated with the expression of its catalytic subunit, human telomerase reverse transcriptase ( hTERT). A correlation of transcript expression with a poor prognosis has been detected in different human malignancies. However, data on hTERT in pancreatic ductal adenocarcinoma (PDAC) are purely descriptive so far. Therefore, we evaluated the impact of hTERT expression on patients' prognosis. Human telomerase reverse transcriptase mRNA isolates from 56 human microdissected PDAC tissues were analyzed by quantitative reverse transcription-polymerase chain reaction and multivariate Cox regression hazard test. Elevated hTERT transcript levels were measured in 23 of 56 PDAC tissues, 33 patients showed no detectable transcripts. Unexpectedly, a low expression of hTERT mRNA levels was associated with a worse prognosis for overall survival ( relative risk = 5.33; P = .013) when compared to high levels, whereas undetectable expression showed an intermediate risk of tumor-related death. These data challenge previous findings outlining hTERT's negative impact on overall survival. The risk pattern obtained in PDAC suggests a more complex regulation of hTERT.

Immune Cell-Associated Protein Expression Helps to Predict Survival in Muscle-Invasive Urothelial Bladder Cancer Patients after Radical Cystectomy and Optional Adjuvant Chemotherapy

Helge Taubert, Markus Eckstein, Elena Epple, Rudolf Jung, Katrin Weigelt, Verena Lieb, Danijel Sikic, Robert Stoehr, Carol Geppert, Veronika Weyerer, Simone Bertz, Astrid Kehlen, Arndt Hartmann, Bernd Wullich, Sven Wach

Summary: Adjuvant chemotherapy is commonly used for muscle invasive bladder cancer patients, but existing biomarkers cannot predict which patients will benefit. Examining immune cell markers, this study found that the presence of tumor-infiltrating immune cells is associated with prognosis, and the impact of chemotherapy on prognosis depends on the expression of immune markers. Identifying specific immune markers, such as macrophages, may help predict chemotherapy response and patient outcomes after radical treatment for bladder cancer.

Integration of Spatial PD-L1 Expression with the Tumor Immune Microenvironment Outperforms Standard PD-L1 Scoring in Outcome Prediction of Urothelial Cancer Patients

Summary: This study highlights the limitations of current PD-L1 scoring algorithms in predicting immune therapy response in urothelial cancer patients. Comprehensive integration of PD-L1 expression and immune phenotypes is suggested for better patient stratification and prediction of treatment outcomes.

GP88/PGRN Serum Levels Are Associated with Prognosis for Oral Squamous Cell Carcinoma Patients

Thomas Greither, Tina Steiner, Matthias Bache, Ginette Serrero, Sven Otto, Helge Taubert, Alexander W. Eckert, Matthias Kappler

Summary: The study suggests that increased levels of GP88 in the serum of oral squamous cell carcinoma (OSCC) patients are associated with a better prognosis, and may serve as an independent predictor of OSCC patients' outcomes. Low serum GP88 levels are significantly linked to a worsened prognosis for OSCC, especially in patients over the age of 60.5.

Combination of GP88 Expression in Tumor Cells and Tumor-Infiltrating Immune Cells Is an Independent Prognostic Factor for Bladder Cancer Patients

Summary: In urothelial bladder cancer, positive expression of GP88 in tumor cells is associated with poor prognosis, while negative expression of GP88 in immune cells is an independent negative predictor. The combination of GP88 staining in both tumor cells and immune cells is a significant independent prognostic biomarker in muscle-invasive BCa.

High Androgen Receptor mRNA Expression Is Associated with Improved Outcome in Patients with High-Risk Non-Muscle-Invasive Bladder Cancer

Summary: High AR mRNA expression is correlated with KRT5 mRNA expression and associated with an improved outcome in high-risk NMIBC, but these molecular markers do not have advantages as independent prognostic factors.

Cell-Free DNA Variant Sequencing Using Plasma and AR-V7 Testing of Circulating Tumor Cells in Prostate Cancer Patients

Verena Lieb, Amer Abdulrahman, Katrin Weigelt, Siegfried Hauch, Michael Gombert, Juan Guzman, Laura Bellut, Peter J. Goebell, Robert Stohr, Arndt Hartmann, Bernd Wullich, Helge Taubert, Sven Wach

Summary: Prostate cancer is a significant cause of morbidity and mortality among men globally, and there is a critical need for biomarkers for diagnosis, prognosis, and therapy prediction. Liquid biopsies like cfDNA and CTCs have shown promise in providing valuable information for prostate cancer patients. In this study, the analysis of cfDNA and CTCs revealed associations with overall survival, disease-specific survival, time to treatment change, and specific gene variants like BRCA1 and AR ligand-binding domain, highlighting their potential in guiding treatment decisions for advanced prostate cancer patients.

Reduced Recurrence Rates Are Associated with Photodynamic Diagnostics Compared to White Light after Extended Transurethral Resection of Bladder Tumors

Alexander Marquardt, Mario Richterstetter, Helge Taubert, Arndt Hartmann, Bernd Wullich, Verena Lieb, Laura Bellut, Sven Wach, Hendrik Apel

Summary: The use of photodynamic diagnostics (PDD) in bladder cancer treatment leads to improved tumor detection rates and reduced recurrence rates. It also reduces the incidence of residual tumors at the initial resection site in extended transurethral resection of the bladder tumor (TURBT) procedures.

Combined miR-486 and GP88 (Progranulin) Serum Levels Are Suggested as Supportive Biomarkers for Therapy Decision in Elderly Prostate Cancer Patients

Alexander Fichte, Angela Neumann, Katrin Weigelt, Juan Guzman, Thilo Jansen, Julia Keinert, Ginette Serrero, Binbin Yue, Robert Stoehr, Thomas Greither, Arndt Hartmann, Bernd Wullich, Helge Taubert, Sven Wach, Verena Lieb

Summary: Our study aimed to evaluate the potential of using miR-486 and GP88 as biomarkers for diagnosing and predicting prostate cancer. The results showed that miR-486 and GP88 were not associated with cancer diagnosis, but significant differences were observed in elderly PCa patients. Additionally, adding miR-486 and GP88 to the baseline model improved the accuracy of predicting definitive therapy.

Genome-Wide Association Screening Determines Peripheral Players in Male Fertility Maintenance

Thomas Greither, Hermann M. Behre, Holger Herlyn

Summary: Deciphering the functional relationships between genes associated with phenotypic variations can be challenging. In this study, a genome-wide association study (GWAS) linked twelve non-synonymous single-nucleotide polymorphisms (SNPs) to fertility variation in men. These SNPs were located in nine genes that have been previously implicated in male fertility. The study also identified potential biomarkers for the diagnosis of male fertility.
