Unimpaired Responses to Vaccination With Protein Antigen Plus Adjuvant in Mice With Kit-Independent Mast Cell Deficiency

Innate inflammatory responses are crucial for induction and regulation of T cell and antibody responses. Mast cell (MC)-deficient Kit mutant mice showed impaired adaptive immunity, suggesting that MCs provide essential adjuvant activities, and pharmacological MC activation was proposed as a new adjuvant principle. However, the Kit mutations result in complex alterations of the immune system in addition to MC deficiency. We revisited the role of MCs in vaccination responses using Mcpt5-Cre R26(DTA/DTA) and Cpa3(Cre/+) mice that lack connective tissue MCs or all MCs, respectively, but feature an otherwise normal immune system. These animals showed no impairment of T and B cell responses to intradermal vaccination with protein antigen plus complete Freund's adjuvant. Moreover, we demonstrate that the adjuvant effects of the MC secretagogue c48/80 in intradermal or mucosal immunization are independent of the presence of MCs. We hence find no evidence for a regulation by MCs of adaptive immune responses to protein antigens. The finding that immunological MC functions differ from those suggested by experiments in Kit mutants, emphasizes the importance of rigorous tests in Kit-independent MC-deficiency models.

Mast Cells in the Skin: Defenders of Integrity or Offenders in Inflammation?

Summary: Mast cells are key effector cells in immediate-type allergic reactions, but they also play an important role in the first-line defense against pathogens, although this can lead to detrimental overshooting responses and exacerbation of inflammatory disorders. In the skin, mast cells have both beneficial functions in maintaining skin barrier integrity and host defense mechanisms, as well as causing inflammatory skin disorders, with their IgE-independent activation pathways playing a crucial role in sustained chronic skin inflammation and disease exacerbation.

Summary: The onset of agriculture around 12,000 years ago had a significant impact on human culture, biology, and health. This study examines the variation in human stature as a proxy for health across a transition to agriculture, and finds that individuals from the Neolithic period were shorter than those from the Upper Paleolithic and Mesolithic. However, the stature gradually increased across subsequent ages. These findings highlight the potential of using different datasets to explore health proxies in prehistory.

Summary: NF-KB signaling pathway plays important roles in regulating cell function and immune response. In this study, we identified B cell defect and dysregulation of immunoglobulin levels in the small intestinal mucosa of Nfkb2-/- mice using transcriptomic and proteomic analysis. NF-KB2/p52 deficiency not only increases resistance to LPS-induced cell apoptosis in the small intestine, but also regulates the plasma cell population and immunoglobulin levels within the gut.

Summary: Organ Mapping Antibody Panels are standardized antibody panels created by the community for multiplexed spatial imaging. This imaging technique allows detailed characterization of molecular and cellular organization in tissues. Although high-parameter data collection is now possible (>60 targets), constructing the necessary antibody panels requires expertise and capital. Organ Mapping Antibody Panels are community-validated resources that save time and money, enhance reproducibility, expedite discovery, and support the creation of a Human Reference Atlas.
