Freshwater Ascomycetes: Jahnula purpurea (Jahnulales, Dothideomycetes), a new species on submerged wood from Martinique Island, Lesser Antilles

Jahnula purpurea J. Fourn., Raja & Shearer, a new species in the Jahnulales (Dothideomycetes) collected from submerged wood in a freshwater river in Martinique Island, Lesser Antilles, is described and illustrated. The characteristic features of the new species are: globose to subglobose, brownish black ascomata with broad, golden brown, subtending hyphae which stain the underlying wood purple; a peridial wall composed of large pseudoparenchymatic cells, which are textura angularis to prismatica in surface view; sparsely septate pseudoparaphyses embedded in a gel matrix; clavate to obclavate asci with a short pedicel; brown, one-septate, ellipsoidal, rough-walled ascospores without a gelatinous sheath or appendages. Unfortunately, because limited material was available from the type collection, we were unable to obtain molecular data. Jahnula purpurea is distinct from all previously described species of Jahnula in its ability to stain the wood purple and in a combination of ascomal, ascus, and ascospore size and morphology.

Freshwater Fungi as a Source of Chemical Diversity: A Review

Summary: This review summarizes the chemical diversity and biological activity of 199 out of 280 known freshwater fungal metabolites published from 1992 to 2020, representing at least seven structural classes. In addition to describing the metabolites themselves, strategies for the collection, isolation, and identification of fungi from freshwater habitats are discussed to enhance chemists' knowledge of mycological principles. The review is expected to serve as a springboard for future natural products studies from the underexplored group of Ascomycota.

Cytotoxic and antimicrobial drimane meroterpenoids from a fungus of the Stictidaceae (Ostropales, Ascomycota)

Summary: New bioactive fungal metabolites dasyscyphins F and G were isolated from a fungus of the Stictidaceae, along with the known dasyscyphin C. The structures were characterized, and a revision was made to the structure of dasyscyphin C based on NMR data. The compounds showed moderate cytotoxicity and antimicrobial activity.

Structural Diversity of Perylenequinones Is Driven by Their Redox Behavior

Summary: This study isolated and characterized the long-postulated biosynthetic precursor of class B fungal perylenequinones from a Shiraia-like sp. Furthermore, it investigated the structural relationship and redox behaviors of hypocrellins and hypomycins. The study found that hypocrellins can undergo reversible reduction and reoxidation under aerobic conditions, and can undergo ring closing metathesis under anaerobic conditions.

Fungal-fungal co-culture: a primer for generating chemical diversity

Genomic and Phenotypic Trait Variation of the Opportunistic Human Pathogen Aspergillus flavus and Its Close Relatives

Summary: This study characterizes the genomic, chemical, and phenotypic variation between strains of pathogenic Aspergillus flavus and its nonpathogenic relatives. The findings reveal extensive heterogeneity within both the pathogenic A. flavus and its close relatives, indicating that strain-level differences may play a major role in the ability of these fungi to cause disease. Surprisingly, strains from nonpathogenic species exhibit similar virulence to A. flavus, challenging the current perception of these species' clinical relevance.

Embellicines C-E: Macrocyclic Alkaloids with a Cyclopenta[b]fluorene Ring System from the Fungus Sarocladium sp.

Summary: This article reports a relatively young class of fungal metabolites, macrocyclic alkaloids with a cyclopenta[b]fluorene ring system, which were first discovered in 2013. The structures of these compounds were identified by analyzing high-resolution electrospray ionization mass spectrometry data and one-dimensional and two-dimensional NMR spectra. The relative and absolute configurations of these alkaloids were determined through various spectroscopic techniques. These compounds showed cytotoxic activity against human cancer cell lines.

Correlative metabologenomics of 110 fungi reveals metabolite-gene cluster pairs

Summary: Natural products research increasingly applies -omics technologies to guide molecular discovery. While the combined analysis of genomic and metabolomic datasets has proved valuable for identifying natural products and their biosynthetic gene clusters (BGCs) in bacteria, this integrated approach lacks application to fungi. The creation of a linked genomics-metabolomics dataset for 110 Ascomycetes and the optimization of gene cluster family (GCF) networking parameters and correlation-based scoring allowed for pairing fungal natural products with their BGCs. The platform identified associations between various natural products and validated BGCs, uncovering new pathways for future discovery.

Summary: Due to resistance, Acinetobacter baumannii is a top priority for drug development according to the World Health Organization. A library of extracts from 2500 fungi was screened for antimicrobial activity against a drug-resistant strain of A. baumannii, and an extract from the fungus Tolypocladium sp. was found to be the most potent. The extract produced pyridoxatin, which may serve as a lead compound for the development of antimicrobials against A. baumannii.

Trachymyrmex septentrionalis ants promote fungus garden hygiene using Trichoderma-derived metabolite cues

Summary: This study reveals that Trichoderma spp. can act as previously unrecognized pathogens of Trachymyrmex septentrionalis fungus gardens. The ants detect and respond to Trichoderma infections through specific secondary metabolites called peptaibols. This discovery is important for understanding ant fungiculture behavior and the protective mechanisms of fungus gardens.
