Summary: This article aims to analyze the baseline distribution of TRPA1, TRPV1, TRPV4, and TRPM8 channels in human systems at the transcriptional level. Using the RNA-seq dataset, the transcriptional levels of these channels were investigated and compared in 95 human subjects representing 33 different tissues, providing essential suggestions for further studies on these proteins.

Summary: Chemotherapy-induced neuropathic pain is a disabling complication that affects patients' quality of life. This study found that bergenin effectively reduces neuropathic pain by modulating TRPA1/TRPV1/NR2B signaling pathway. The findings suggest that bergenin could be a promising therapeutic approach for chemotherapy-induced neuropathic pain.

Summary: Musculoskeletal pain, especially after traumatic muscle injury, is a common and poorly managed public healthcare issue. This study found that the transient receptor potential ankyrin 1 (TRPA1) may be a promising anti-inflammatory and antinociceptive target in treating pain after traumatic muscle injury.

Summary: Burn injuries are often overlooked, but they have significant impacts on health and can even be fatal. Overexposure to UV radiation can cause serious clinical effects and is a major public health concern. Recent studies have made progress in understanding the molecular aspects of sunburn caused by UVB radiation, specifically focusing on the role of TRPA1 and TRPV1 channels in modulating inflammatory, oxidative, and proliferative processes. However, the mechanisms underlying the activation of these channels in sunburn induced by UVB radiation are still unknown.

Summary: Persistent reprogramming of epigenetic pattern leads to changes in gene expression in neurological disorders. TRPA1, a member of TRP channels superfamily, is activated by migraine triggers, expressed in trigeminal neurons and brain regions important in migraine pathogenesis, and regulates the epigenetic profile of pain-related genes. Epigenetic regulation of TRPA1 may play a role in the efficacy and safety of anti-migraine therapies targeting TRP channels and CGRP. TRPA1 is also involved in neurogenic inflammation, important in migraine pathogenesis. Further exploration of the epigenetic connections of TRPA1 is necessary for efficient and safe anti-migraine treatment.

Summary: Inflammation and oxidative stress are processes associated with various diseases. This study investigates the potential of CiL-1, a compound found in seaweed, as an anti-nociceptive and anti-inflammatory agent. Results show that CiL-1 effectively reduces pain and inflammation in adult zebrafish. Additionally, CiL-1 reduces the levels of reactive oxygen species (ROS) in the brain and liver. In silico analyses suggest that CiL-1 interacts with specific ligands and may modulate the TRPA1 channel, explaining its anti-inflammatory effects.

Summary: The study systematically evaluated the concentration-dependent effects of NO on human HSPC behavior in vitro, revealing a biphasic role where low concentrations activated NO signaling and promoted proliferation, while high concentrations slowed proliferation and induced differentiation. This highlights the importance of using appropriate NO concentrations in in vitro cultures of HSPCs, with potential implications for clinical application in cellular therapies.

Summary: This study investigated the involvement of TRPV1 in nitric oxide-mediated influence on hippocampal processes. The findings suggest that TRPV1 channels can be activated to modulate excitatory neurotransmission in the hippocampus, and this effect is linked to nitric oxide production.

Summary: TRPA1 is a member of the TRP superfamily of cation channels and is widely expressed in sensory neural pathways. It detects irritant chemicals, hypoxia, and hyperoxia. Its role in respiratory and behavioral modulation has been studied using Trpa1 knockout mice. The results show that TRPA1 is necessary for protective responses in respiration and behavior, suggesting that TRPA1 channels in the airway may play a sentinel role for environmental threats.
