Summary: Inhibition of cathepsin K enhances oxaliplatin-induced apoptosis by increasing OTUB1 phosphorylation, promoting p53 stabilization, and upregulating Bax expression, leading to decreased tumor size. There is a strong correlation between phosphorylation of OTUB1(Ser16) and p53/Bax expression in human renal clear cell carcinoma tissues.

Summary: The study evaluated the activation of p53 and p27 pathways for attenuating hepatocellular carcinoma. A novel compound, hyrtiosone A, isolated from a marine demosponge, exhibited potential anti-inflammatory and antioxidant properties, making it a promising candidate for liver cancer treatment. The compound showed in vitro anticancer activity in HepG2 cells by inducing apoptosis and cell cycle arrest, and upregulating p53 and p27 gene expression.

Summary: The study reveals that IFI16 and the p53 pathway activator nutlin-3 synergistically induce apoptosis in U2OS and A549 cells, and phosphorylation of p53 at serine 392 by protein kinase R plays a critical role in IFI16-p53-dependent apoptosis. Overexpression of STING suppresses p53 serine 392 phosphorylation, p53 transcriptional activity, and p53-dependent mitochondrial depolarization and apoptosis, indicating a protumorigenic role for STING in certain cancer types by degrading upstream IFI16.

Summary: This study demonstrates that oxaliplatin can induce cellular senescence in colorectal cancer cells, particularly in those with p53 proficiency and p14(ARF) expression. The induction of senescence by oxaliplatin is associated with G2-phase arrest and activation of p53/p21(CIP1) pathway, highlighting the importance of these factors in cytostatic drug-based tumor therapy.

Summary: Merosesquiterpenes show promise as potential therapeutic agents for colorectal cancer, as they induce DNA damage, trigger apoptosis, and exhibit cytotoxic activity against CRC cell lines and tumor organoids. Their efficacy is observed in both wild-type and mutant p53 CRC cells, making them a potential candidate for CRC chemotherapy.

Summary: This review discusses the role of the p53 gene in regulating ferroptosis, primarily through influencing metabolic networks and signaling pathways that affect tumor cell sensitivity to ferroptosis. This has important implications for further understanding the role of p53 in tumor ferroptosis and developing new strategies for cancer treatment.

Summary: Disruption of circadian rhythms can increase the risk of various cancers. Mutations in the CRY2 gene found in human cancers have been shown to accelerate cell growth in mouse fibroblasts expressing high levels of c-MYC. These mutations also have divergent effects on circadian rhythms and interaction with SCFFBXL3.

Summary: In this study, it was found that miR-3622a-3p inhibited cell migration and invasion in human prostate cancer, while miR-3622b-3p promoted cell proliferation, migration, and invasion. miR-3622 regulated the p53-downstream gene network to control apoptosis and the cell cycle. AIFM2 was identified as a direct target of miR-3622b-3p and responsible for miR-3622 knockout-induced apoptosis. Additionally, miR-3622 knockout inhibited the epithelial-mesenchymal transition involved in prostate cancer metastasis through upregulation of vimentin.

Summary: Autophagy is a crucial biological process for maintaining cell homeostasis, with levels usually lower under normal conditions but still with important regulatory factors present. A study revealed that the autophagy-related protein ATG5 regulates c-Myc protein degradation through the ubiquitin-proteasome pathway in normal conditions, limiting cell growth and playing a key role in embryonic stem cell differentiation.

Summary: Hypoxia is crucial in solid tumors, with HIF and p53 signaling pathways playing key roles in regulating cellular responses to hypoxia. The interplay between hypoxia and p53 pathways can impact cancer progression, with p53 regulating hypoxia and HIF signaling in various ways, while mutant p53 can promote cancer progression through interaction with hypoxia and HIF signaling.

Summary: During oncogenesis, cells undergo unrestricted proliferation, altering tissue homeostasis and possibly due to dysregulation of p53 family proteins p63 and p73. While p63 and p73 can compensate for loss of p53 in some cases, their overlap with p53 functions is strongest in regulating p53-dependent gene expression. Surprisingly, p73 is rarely lost or mutated in cancers, with its inactive isoforms often overexpressed and promoting cancer growth by repressing cell death mediated by p73.

Summary: In addition to mutations in the p53 gene, compromised p53 functions due to decreased nuclear p53 protein levels or enhanced turnover are found in various human cancers, contributing to tumorigenesis. The degradation of p53 by HPV viral E6 protein or growth-promoting signaling also plays a critical role in cancer development. The association between p53 downregulation and carcinogenesis highlights the importance of basally expressed p53 in maintaining cellular homeostasis.

Summary: PLAAT4 is a class II tumor suppressor with phospholipid metabolizing abilities. It functions as a downstream effector of retinoic acid signaling and plays a role in tumor prevention and anti-pathogen defense. Recent research has also identified its anti-microbial function, favoring protection from virus and parasite infections.
