Summary: Metabolic disturbance, particularly of glucose metabolism, is a hallmark of tumors such as non-small cell lung cancer (NSCLC). This review focuses on research advances in glycolysis in NSCLC and systematically provides an overview of the key enzymes, biomarkers, non-coding RNAs, and signaling pathways that modulate the glycolysis process and, consequently, tumor growth and metastasis in NSCLC. Current medications, therapeutic approaches, and natural products that affect glycolysis in NSCLC are also summarized. The development of metabolic intervention that targets glycolysis, alone or in combination with current therapy, is a potential therapeutic approach in NSCLC treatment.

Summary: POLQ, an overexpressed DNA polymerase in non-small-cell lung cancer (NSCLC), is correlated with high tumor stage, poor prognosis, increased tumor mutational burden, and ALK and TP5 mutation status. Inhibition of POLQ impairs lung tumorigenesis and enhances radiosensitivity by prolonging the G2/M phase and promoting faster DNA damage repair. Novobiocin (NVB), a specific POLQ inhibitor, targets cancer cells without causing significant toxicity to normal pulmonary epithelial cells. These findings suggest that POLQ is a promising target for improving radiotherapy efficacy in NSCLC.
