Ptr null mutant embryos displayed severe alterations in axonal morphology, glial cell distribution, and peripheral nervous system defects. Cell-based assays revealed that Ptr interacts directly with Hh and may act as a negative regulator of Hh signaling.

Summary: Clear cell renal cell carcinoma (ccRCC) is the deadliest neoplasm of the urinary tract. The expression of Sonic Hedgehog (SHH) was found to be higher in grade 1 tumors and decreased with tumor progression. PTCH expression was lower in grades 1 and 2 compared to the control and grade 4, while SMO expression was higher in grade 4 than in other grades and the control. High PTCH and low SMO expression were associated with better survival rates in ccRCC patients.

Summary: This study aimed to investigate the anti-cancer effects of berberine on colorectal cancer (CRC) and explore its underlying mechanism involving the Hedgehog signaling pathway. The results showed that berberine suppressed the proliferation, migration, and invasion of CRC cells, induced cell apoptosis, and blocked the cell cycle. Additionally, berberine inhibited tumor growth and promoted apoptosis and cell cycle arrest in a mouse model of CRC. However, the potential adverse reactions of berberine should be considered when using it.

Summary: This study investigated the role of the Hedgehog (Hh) pathway in modulating the immune response. The results showed that Hh pathway activation favored the Th2 phenotype and impaired the establishment of the Th1 phenotype. Additionally, mice with constitutive activation of the Hh pathway showed a milder response to inflammatory challenge and less spinal cord damage. These findings demonstrate the involvement of the Hh/ptch pathway in immune response modulation and highlight the potential of the ptch(+/-) mouse model for studying T-cell-mediated diseases and developing new therapeutic strategies for inflammatory diseases.
