Summary: Human cytomegalovirus (HCMV) infection in fetal brains can cause neurodevelopmental disorders by targeting neural progenitor cells (NPCs). This study investigated the expression of cellular and viral microRNAs (miRNAs) in HCMV-infected NPCs during early infection. Several differentially expressed cellular miRNAs and upregulated HCMV miRNAs were identified. Deep learning methods were used to predict miRNA targets and their associations with cellular mRNA expression changes. The study revealed the involvement of these miRNAs in neurodevelopmental and cell-cycle processes, and confirmed the regulatory role of hcmv-miR-US25-1 on the Jag1 gene, an important factor for neurogenesis.

Summary: The study investigated the role of CDC25C in neural progenitor cell division and differentiation by knocking down the gene in developing rat brains. The results showed that CDC25C is essential for maintaining the proliferative state of neural progenitor cells during cortical development. Knockdown of CDC25C resulted in early cell cycle exit and premature differentiation of neural progenitor cells, revealing a novel role for CDC25C in NPC division and cell fate determination.

Summary: Mesenchymal stem cells (MSCs) have been used in clinical studies due to their versatility and low immunogenicity. However, safety issues remain a concern. Recently, extracellular vesicles (EVs) derived from MSCs have been explored as an alternative. EVs are nano-sized particles released by cells and have similar biological functions as their origin cells. They carry bioactive molecules and facilitate tissue regeneration. EVs derived from adipose-derived MSCs (ADMSC) also have neuroprotective and neurogenesis effects. This study aimed to enhance the neural differentiation ability of ADMSC-derived EVs by investigating the neurogenic-differentiation process. The results showed that differentiated EVs (dEVs) obtained from neurogenesis conditioned media increased neurogenic ability by altering microRNA expression and cytokine composition. dEVs promoted neuronal differentiation of neural progenitor cells in vitro, indicating their potential in EV-based therapy for neurological disorders.

Summary: By performing RNAseq transcriptomic profiling of 110 patient samples, we revealed a disease continuum from normal sun-exposed skin to precancerous actinic keratosis (AK), and then to malignant invasive cutaneous squamous cell carcinoma (cSCC) and potentially lethal metastatic disease. This progression is accompanied by the suppression of master regulators of epidermal differentiation, dynamic modulation of the epidermal differentiation complex, remodelling of the immune landscape, and an increase in tumor specific keratinocytes. Our study provides a comprehensive map of the cSCC disease continuum and reveals potentially actionable events that promote and accompany disease progression.

Summary: Neurogenesis is a complex process that relies on the balanced execution of expression programs involving various molecular players, including transcription factors, chromatin modulators, and noncoding RNAs. In this study, the function of the long noncoding RNA (lncRNA) Malat1 was investigated during neural embryonic stem cell (ESC) differentiation. Deletion of Malat1 inhibited the proliferation of neural progenitor cells (NPCs) and resulted in increased expression of miR-26a and miR-26b. Inactivation of miR-26a/b rescued the proliferative phenotype of Malat1 knockout cells and accelerated neuronal differentiation. This study reveals an unknown interaction between Malat1 and miR-26 in the regulation of NPC proliferation and neuronal differentiation.
