Alpha1-acid glycoprotein is contained in bovine neutrophil granules and released after activation

The present study was designed to investigate the capability of bovine neutrophil granulocytes to produce the minor acute phase protein alpha(1)-acid glycoprotein (AGP, Orososmucoid). Bovine neutrophils contain a high MW (50-60 kDa) AGP isoform (PMN-AGP), as determined by Western blotting and confirmed by fluorescence microscopy. The presence of AGP in bovine neutrophils has been confirmed by fluorescence immunocytometry. In addition, bovine neutrophils contain also a 42-45 kDa isoform, which has the same MW as plasma-, liver-delivered, AGP. cDNA sequence of plasma- and PMN-AGP revealed that (i) the two proteins are products of the same gene; (ii) the differences in molecular weight are due do different post-translational modifications. This result was confirmed by deglycosylation of the two glycoforms. Exocytosis studies showed that isolated neutrophils exposed to several challengers, including Zymosan activated serum (ZAS) and phorbol 12-myristate 13-acetate (PMA), which mimic the inflammatory activation, released PMN-AGP as early as 15 min. AGP's mRNA is physiologically expressed by mature resting neutrophils. Real-time PCR on LPS, ZAS and PMA challenged cells revealed that the level of expression apparently does not increase after inflammatory activation. Collectively, the findings reported in this paper proved that PMN-AGP: (i) is a hyperglycosylated glycoform of plasma AGP, (ii) is stored in granules, and (iii) is released by neutrophils in response to activation. Due to its anti-inflammatory activity, PMN-AGP may work as a fine tuning of the neutrophils functions in the inflammatory focus, i.e. it can reduce the damages caused by an excess of inflammatory response. Summary: This preliminary pilot study found that high levels of miR-30b-5p are associated with milder forms of myxomatous mitral valve disease in Cavalier King Charles Spaniels. However, the results did not show statistical significance between miR-30b-5p expression and heart murmur intensity, mitral regurgitation severity, and Mitral INsufficiency Echocardiographic score. The study also demonstrated a relationship between high abundance of miR-30b-5p and echocardiographically more stable forms of the disease over time.

A refined culture system of oocytes from early antral follicles promotes oocyte maturation and embryo development in cattle

Summary: The aim of this study was to improve the in vitro culture of bovine oocytes collected from early antral follicles (EAFs) by maintaining physiological conditions and adding key hormones. The culture system involved a 5-day culture period and 3D-like structures. The cultured oocytes showed morpho-functional changes, improved developmental competence, and potential for in vitro embryo production.

CPEB1-dependent disruption of the mRNA translation program in oocytes during maternal aging

Summary: In the absence of transcription, gene expression required for oocyte maturation is regulated by translation of preexisting mRNAs. A defective program of mRNA translation in oocyte is associated with maternal aging, leading to a decline in oocyte quality during aging. Dysfunction in the oocyte translation program is characterized by disrupted ribosome loading, altered translation of 3'-UTR-reporters, altered poly(A) length of mRNAs, decreased levels of Cpeb1 mRNA translation and protein, and de-repression of Ccnb1 translation in old oocytes.

Summary: This study developed a fast and easy protocol for isolating small extracellular vesicles (sEV) from plasma of mast cell tumor (MCT)-affected dogs. It was found that sEV-miR-21-5p, a miRNA overexpressed in MCT, was significantly associated with sEV in dogs with nodal metastasis. These findings provide valuable insights into the role of sEV in disease pathogenesis.

Cotinine as a Sentinel of Canine Exposure to Tobacco Smoke

Summary: This study aimed to measure cotinine concentration in the serum and hair of dogs that were exposed to the owner's tobacco smoke and to compare it with that of unexposed dogs. The results showed that cotinine concentrations were significantly higher in exposed dogs than unexposed dogs, and there were differences in cotinine concentrations between different sex dogs. These results confirmed cotinine as a marker of passive smoking in dogs.

Association between Mastitis Occurrence in Dairy Cows and Bedding Characteristics of Compost-Bedded Pack Barns

Tobacco Smoke Exposure in Pregnant Dogs: Maternal and Newborn Cotinine Levels: A Pilot Study

However, the risks for pets, especially dogs, have been largely overlooked. A recent study found that cotinine, a primary metabolite of nicotine, can be detected in dogs living with smoking owners, indicating the potential adverse health consequences for pets. This highlights the importance of raising awareness among pet owners about the risks of smoke exposure.

Review: The putative role of Progesterone Receptor membrane Component 1 in bovine oocyte development and competence

Summary: PGRMC1 is expressed in multiple cell types of the mammalian reproductive system and plays a role in follicular growth in the ovary and proper completion of oocyte maturation. It is also involved in mitotic and meiotic cell division through its association with the spindle apparatus and chromosomal passenger complex. The role of PGRMC1 in preimplantation embryo is still unclear and further research is needed.
