Background: Rodent models are a very helpful tool to investigate immunological mechanisms in allograft rejection. The aim of this study was to compare two different immunodeficient recipients in a humanized mouse model of arterial xenotransplantation in terms of reconstitution of the human immune system and rejection of the arterial graft. Methods: Side branches of human mammary artery were transplanted as infrarenal aortic interposition grafts into C.B-17-SCID beige and C57BL/6-Rag2(-/-)gamma c(-/-) recipients. 7 days after surgery mice were reconstituted with 5 x 10(7) human peripheral blood mononuclear cells (hu PBMCs) and 30 days after reconstitution mice were sacrificed and histologic analysis was performed. Peripheral blood and splenocytes were investigated by FACS and ELISA analysis to ensure engraftment of human CD45(+) cells. Results: Transplant arteriosclerosis developed in non-PBMC-reconstituted C.B-17-SCID beige mice (intimal proliferation: 36.31 +/- 4.37%), but significantly less in C57BL/6-Rag2(-/-) gamma c(-/-) recipients (intimal proliferation: 12.26 +/- 5.21%). After reconstitution with 5 x 10(7) unfractionated human PBMCs both mouse strains showed intima proliferation 30 days after reconstitution (C.B-17-SCID beige: 28.49 +/- 7.95% and C57BL/6-Rag2(-/-) gamma c(-/-): 44.58 +/- 11.08%). Whereas only very few human CD45+ cells were found in mouse blood and spleen of C.B-17-SCID beige mice, C57BL/6-Rag2(-/-) gamma c(-/-) mice revealed a reliable reconstitution. In addition, levels of human IgG and IgM within the peripheral blood were markedly higher in C57BL/6-Rag2(-/-) gamma c(-/-) recipients. Conclusion: In this study we can show, that the use of C57BL/6-Rag2(-/-) gamma c(-/-) mice may be advantageous compared to C.B-17-SCID beige recipients in a humanized mouse model of vessel transplantation. Summary: Chronic dialysis patients with AS undergoing TAVI or SAVR had similar 1-year mortality rates, with better survival at 30 days for TAVI patients. TAVI may improve peri-procedural outcomes, although TAVI patients required more pacemaker implantation and showed more aortic regurgitation, while SAVR patients required more blood transfusions and had longer hospital stays.

Summary: Higher CKD stages were associated with lower survival rates for patients undergoing both TAVI and SAVR. CKD >= 3b and CKD >= 3a were identified as the major independent risk factors for mortality in TAVI and SAVR patients, respectively. Stratification based on CKD substage may help in selecting appropriate patients for these therapies.

Outcomes of aortic valve repair: early results from the German Aortic Valve Registry

Summary: AV repair/AV-sparing root surgery is an effective strategy for treating young patients with aortic regurgitation and/or aortic root aneurysm. This study analyzed real-world data from the German Aortic Valve Registry and found that the surgical procedure resulted in satisfactory clinical outcomes, including high survival rates and freedom from cardiac adverse events at one year. Additionally, the specific surgical technique used did not significantly impact the outcomes.

Cross-Species Analysis of Innate Immune Antagonism by Cytomegalovirus IE1 Protein

Summary: The study analyzed the functions of human cytomegalovirus (CMV) immediate early 1 (IE1) protein and rat CMV IE1 (rIE1) protein, and found that their interactions with cellular proteins PML and FEN1 were species-specific. It was also discovered that both IE1 proteins can antagonize human STAT2, but different regions of the viral proteins were involved in the interaction. Additionally, the study revealed that the binding of PML, FEN1, and STAT2 was conserved between primate and rodent proteins, but rIE1 lacked chromatin tethering function.

Inhibitors of Activin Receptor-like Kinase 5 Interfere with SARS-CoV-2 S-Protein Processing and Spike-Mediated Cell Fusion via Attenuation of Furin Expression

Summary: Screening of a protein kinase inhibitor library identified SB431542 as a compound that interferes with SARS-CoV-2 replication by targeting activin receptor-like kinase 5 (ALK5). ALK5 inhibition downregulates furin expression induced by TGF-beta and reduces spike-mediated cell-cell fusion, thereby attenuating SARS-CoV-2 infectivity and cell-cell spread. This suggests that ALK5 inhibitors may be a potential therapy option for COVID-19, especially in cases with TGF-beta dominated cytokine storm.

Ex vivo evaluation of the Ozaki procedure in comparison with the native aortic valve and prosthetic valves

Summary: In this ex vivo study, the hydrodynamic performance and cusp kinematics of the Ozaki neocuspidized aortic valve were compared with native aortic and prosthetic valves. The results showed that the Ozaki valve had excellent hydrodynamic performance and similar cusp motion characteristics to the native aortic valve, suggesting potential beneficial clinical outcomes.
