Summary: Treprostinil significantly reduced peak elevated serum creatinine levels and accelerated normalization relative to IRI-placebo. Treatment with treprostinil inhibited IRI-mediated renal apoptosis, mitochondrial oxidative injury, and the release of cytochrome c versus IRI-placebo. Overall, treprostinil reduced mitochondrial-mediated renal apoptosis, inhibited mitochondria fission, and promoted mitochondria fusion, thereby accelerating mitochondrial recovery and protecting renal proximal tubules from renal IRI.

Summary: In this study, we evaluated the dynamic changes in autophagy during IgAN. We found that autophagy was persistently induced in IgAN rats and the expression level of inflammation was also elevated. The use of the autophagy inhibitor 3-methyladenine alleviated renal injury and inflammation in IgAN rats. These findings provide further evidence for autophagy as a therapeutic target for IgAN.

Summary: In malignant hypertension, the most severe hypertension-induced kidney injury occurs. In a rat model of malignant hypertension, a broad stimulation of inflammatory mechanisms was observed, but specific pathways, such as activation of the complement cascades, were activated only in the malignant form of the disease. Complement inhibitors may alleviate the thrombotic microangiopathy of malignant hypertension even in the absence of primary complement abnormalities.
