Summary: This study presents a new conditional knockout strategy that enables efficient counter selection in mouse embryonic stem cells. The strategy overwrites counter selectable markers and allows the removal or replacement of wild-type DNA with the desired payload DNA. The restored native gene function in the conditional knockout cells facilitates subsequent mouse development and the generality of the strategy is demonstrated through the knockout of other genes.

Summary: Traditional therapies for male infertility focus on restoring fertility or preserving sperm in severe cases, but have limitations that impact their effectiveness. Reproductive medicine has turned to biotechnological alternatives, such as tissue-engineered testicular tissue reconstruction, to improve infertility treatment and gamete preservation. This approach mimics the testicular microenvironment to maintain male gametes in culture or produce viable grafts for transplantation. The use of biomaterials plays a crucial role in these artificial biological systems, but each material has advantages and disadvantages. This review discusses the progress and challenges in testicular regenerative medicine and the reconstruction of the testicular microenvironment using tissue bioengineering approaches.

Summary: Nickel chloride has adverse effects on reproductive function in male mice, including reducing sperm concentration and motility while increasing sperm malformation rates. It also causes damage to testis and epididymis tissues, decreases serum testosterone levels, and down-regulates the expression of certain testis-related genes. These findings suggest that nickel chloride disrupts spermatogenesis through testicular damage and disturbance of the hypothalamic-pituitary-testis axis.

Summary: There is a significant demand for safe and effective non-hormonal male contraceptives, but research on male contraceptive drugs has lagged behind that of female birth control pills. However, a new potential male contraceptive candidate, BHD, has been developed through the design and synthesis of a new series of molecules derived from lonidamine. BHD demonstrated a 100% contraceptive effect on male mice and rats, reducing fertility to 90% and 50% respectively after 6 weeks of treatment. BHD also induced rapid apoptosis of spermatogenic cells and effectively disrupted the blood-testis barrier.

Summary: This study aims to clarify the expression of PRDX2 in the testes of obese mice and lay a foundation for further exploration of its regulatory and protective effects on spermatogenesis. The results showed that PRDX2 expression was reduced in the testes of obese mice, and hydrogen peroxide and cholesterol had different regulatory effects on the expression of PRDX2 in Sertoli cells.

Research using the Acrv1 gene revealed that the gene's proximal promoter contains necessary cis-regulatory sequences, an insulator prevents somatic cell expression, RNA II polymerase is paused in spermatocytes, and TDP-43 maintains the paused state. However, the transcription factor responsible for round spermatid-specific transcription remains elusive.
