Antioxidant activity of whey protein fractions isolated by gel exclusion chromatography an protease treatment

Whey proteins were isolated from whey powder by a combination of gel exclusion chromatography and protease (pepsin or trypsin) treatment. Whey solution (6 g/dl) was applied to Sephadex G-200 column chromatography and three fractions were obtained. Gel electrophoresis (SDS-PAGE) was used to identify the fractions; the first one contained immunooglobulins and bovine serum albumin, the second contained P-lactoglobulin and a-lactalbumin whereas the third fraction contained small peptides. We have also subjected the whey filtrate to proteases (pepsin and trypsin). Treatment with proteases showed that P-lactoglobulin can be obtained after hydrolysis of the second fraction with pepsin. When the whey filtrate was treated with pepsin and then applied to Sephadex G-200 column chromatography three fractions were obtained; the first one was bovine serum albumin, the second was beta-lactoglobulin and the third fraction contained small peptides. After trypsin treatment only two fractions were obtained; the first one was serum albumin and the second fraction was an alpha-lactalbumin rich fraction. We have determined the antioxidant activity of the fractions using an assay based on the measurement of superoxide radical scavenging activity. Our results showed that among the three fractions, the first fraction had the highest superoxide radical scavenging activity. Also, protease treatment of the second fraction resulted in an increase in the antioxidant activity. (c) 2007 Elsevier B.V. All rights reserved.

Evaluation of oxidative stress biomarkers and antioxidant parameters in allergic asthma patients with different level of asthma control

Summary: This study aimed to evaluate the differences in oxidative stress biomarkers and antioxidant capacity between asthma patients and healthy individuals. The results showed a significant increase in levels of malondialdehyde and protein carbonyls, as well as a decrease in reduced glutathione levels, in asthma patients. These findings suggest the presence of severe oxidative stress in asthma patients even under controlled conditions.

Identification of natural compounds of Jurinea species by LC-HRMS and GC-FID and their bioactivities

Summary: The study aimed to determine the chemical composition and biological activities of ethanol and n-hexane extracts of three different Jurinea species. Results showed that certain extracts exhibited cytotoxic effects on cancer cells and high antioxidant activities. These plants may be considered as alternative sources of cytotoxic and/or antioxidant flavonoids, phenolics, and unsaturated fatty acids.

Summary: This study aimed to compare the levels of oxysterols and lipid peroxidation products in allergic asthma patients and healthy controls, and provide information about the involvement of lipid peroxidation in allergic asthma. The results showed that plasma levels of oxysterols and lipid peroxidation products were significantly higher in allergic asthma patients compared to healthy controls, suggesting their involvement in the pathogenesis of allergic asthma.

Summary: In this study, a fluorescent biosensing system for high sensitivity detection of MT was established using a new Ag+-stabilized DNA triplex probe (Ag+-SDTP) combined with catalytic hairpin assembly (CHA) and CRISPR/Cas12a signal enhancements. The sensing method is able to selectively detect MT from other non-specific molecules and can achieve low level detection of MT in diluted human serums, demonstrating its potential for monitoring disease-specific MT biomarker at trace levels.

Summary: Alzheimer's disease is a major cause of senile dementia and is characterized by the aggregation of neurofibrillary tangles and amyloid plaques in the brain. The A beta 1-42 peptide is considered the primary cause of neurotoxicity due to its insolubility and subsequent aggregation. A beta 1-42 and A beta 1-40 have been identified as relevant biomarkers for AD diagnosis, and SurfaceAssisted Laser Desorption/Ionization Mass Spectrometry (SALDI-MS) was used to monitor the A beta 1-42/A beta 1-40 ratio without prior sample treatment.

CRISPR-empowered electrochemical biosensor for target amplification-free and sensitive detection of miRNA

Summary: We developed a CRISPR-empowered electrochemical biosensor, named PER-E-CRISPR, for amplification-free and sensitive detection of miR-21. The biosensor combines the advantages of CRISPR/Cas13a and primer exchange reaction (PER) for target amplification. Under optimized conditions, the PER-E-CRISPR assay exhibits a linear range from 10-13 to 10-7 M and a limit of detection of 30.2 fM. The biosensor shows excellent performance in actual plasma, indicating promising prospects for miRNA detection in molecular diagnosis.

Application and evaluation of molecular docking for aptamer and small molecular interaction- A case study with tetracycline antibiotics

Summary: Molecular docking (MD) analysis is widely used for studying the interaction between aptamers and small molecules. This study established steady-state aptamers of tertiary structures (SATS) using specific aptamers of tetracycline antibiotics as docking models. Molecular docking results revealed multiple binding sites in the SATS of aptamers, with significant variations in binding free energy (BFE) and docking score (DS). The study also proposed a method for MD analysis based on SATS, providing insights into binding mode and predicting binding sites.

The construction of highly selective surface molecularly imprinted polymers based on Cu(II) coordination for the detection of bisphenol A

Summary: In this study, a highly selective molecularly imprinted polymer Cu-MIPs@CS was designed and synthesized for bisphenol A (BPA). It also exhibited superior discrimination ability between BPA and its structural analogue. Additionally, Cu-MIPs@CS were successfully applied as a solid phase extraction adsorbent for the determination of BPA in drinking water samples.
