Summary: Despite the widespread use of vaccines, Streptococcus pneumoniae remains a leading cause of bacterial pneumonia. Vaccines are effective against most serotypes, but rise in nonvaccine serotypes and moderate efficacy against some vaccine serotypes contribute to high disease incidence. Human monoclonal antibodies targeting conserved pneumococcal proteins show potential for disease prevention and treatment, with opsonophagocytic activity suggesting a mechanism of protection.

This study investigated the use of polymeric nanoparticles to deliver PspA, a candidate for new pneumococcal vaccines, and found that PGA-co-PDL/HCl-CS or PLGA/HCl-CS nanoparticles with encapsulated or adsorbed PspA provided serotype-independent protection.

Summary: Experimental Human Pneumococcal Challenge (EHPC) is a safe procedure for studying vaccine research, with no serious adverse events related to pneumococcal inoculation. Colonised individuals were more likely to experience safety events, but most symptoms were mild and could be managed conservatively. Outpatient human pneumococcal challenge can be conducted safely with appropriate safety monitoring procedures in place.

Summary: A multicentre, retrospective observational study in eight Latin American and Caribbean countries assessed the direct effect of pneumococcal conjugate vaccines (PCVs) on the distribution of pneumococcal serotypes causing invasive pneumococcal disease in children younger than 5 years. The study found a significant decrease in the percentage of vaccine serotypes in PCV countries after PCV introduction.

Summary: The incidence of antibiotic-nonsusceptible invasive pneumococcal disease (NS-IPD) has decreased in the United States after the introduction of 7- and 13-valent pneumococcal conjugate vaccines (PCVs). However, there has been an increase in nonvaccine type NS-IPD, particularly among older adults. The use of higher valency PCVs containing the common nonsusceptible serotypes could help further reduce NS-IPD.

Summary: This study identified genetic variations in pneumococcal carriage that differ between infants and adults. Serotypes and genetic background were found to explain a proportion of the heritability in carriage. Summary: This study assessed the impact of pediatric 13-valent pneumococcal conjugate vaccine (PCV13) on pneumococcal meningitis in adults in Japan from 2014 to 2018. Results showed that younger age and lower case fatality rate were associated with meningitis in adult patients. Factors such as splenic dysfunction, specific serotypes, and age influenced the occurrence of meningitis. PCV13 had an indirect effect on non-meningitis cases, but its impact on meningitis was limited due to an increase in non-PCV13 serotypes.

It was found that patients with Bare Lymphocyte Syndrome (BLS) exhibited clinical findings similar to severe combined immunodeficiency (SCID), and the flow cytometric MHC-II expression study played a crucial role in the diagnosis, differential diagnosis, early hematopoietic stem cell transplantation (HSCT), and post-HSCT follow-up.

Summary: This study demonstrates that IgG seroconversion during male infection can amplify female immunopathology and infertility. IgG transcytosis through the FcRn receptor allows Chlamydia to cross the epithelial barrier and be phagocytosed by antigen-presenting cells. This leads to increased pro-inflammatory signaling and T cell proliferation, resulting in more severe pathology and infertility in female mice.
