Summary: NOD2 deficiency contributes to fibrosis and stricturing complications in Crohn's disease through dysregulated homeostasis of activated fibroblasts and macrophages, which can be ameliorated by gp130 blockade. Carriers of NOD2 risk alleles exhibit dysregulated homeostasis of activated fibroblasts and macrophages, suggesting a potential role for gp130 blockade in rescuing the activated program caused by NOD2 mutations.

Summary: Blau syndrome (BS) is a rare genetic immune disease commonly seen in childhood, with a high miss-rate of diagnosis and lack of effective clinical management. This case report presents a 54-year-old Chinese male patient with hand malformation, fever, skin rash, and joint pain, who was ultimately diagnosed through medical history and genetic analysis. The report aims to raise awareness among clinicians for accurate diagnosis and appropriate treatment of this rare clinical entity.

Summary: Phage therapy may induce an immune response and production of neutralizing antibodies. The presence of these antibodies can affect the success of therapy, but it does not necessarily guarantee failure. On the other hand, phage-antibody complexes may cause tissue damage and late complications, highlighting the importance of monitoring antibody levels and discontinuing therapy if necessary.

Summary: Prediction of endoscopic post-operative recurrence in CD patients following ICR using clinical risk factors alone has been inadequate. This study explored the association and predictive value of PBL DNA methylation in CD patients following ICR. Results showed limited differences in the genome-wide DNA methylome among CD patients with and without endoscopic POR, indicating that PBL DNA methylation is not a feasible predictive tool in post-operative CD.

Summary: This study retrospectively analyzed the safety of robotic redo ileocolonic resection in 98 patients with Crohn's disease. Patients who underwent redo ileocolonic resection had longer disease duration, more associated anoperineal disease, more previous lines of medical treatments, more frequent use of total parental nutrition before surgery for malnutrition correction, and longer adhesiolysis time. The risk of conversion to open ileocolonic resection was higher in the redo group. There were no significant differences in length of stay and 30-day morbidity between the two groups. No mortality or anastomotic leaks occurred in either group.

Summary: Hearing impairment significantly affects communication between cancer patients and their oncologists. A questionnaire was developed with otorhinolaryngologists and hearing-impaired patients, analyzing the experiences, difficulties, wishes, and life satisfaction of 104 cancer survivors. The study found differences in conversation difficulties, physician-patient communication wishes, and psychological well-being between subjectively hearing-impaired and normal hearing participants.
