Summary: The effects of integrated stress response inhibitor (ISRIB) on cellular senescence and pulmonary fibrosis in silicosis were evaluated. ISRIB significantly reduced the degree of pulmonary fibrosis and reversed the expression of senescence-related factors in mice with silicosis. These findings suggest that ISRIB may alleviate pulmonary fibrosis in silicosis by inhibiting cellular senescence.

Summary: In this study, mannose-modified hierarchically porous UiO-66 nanoparticles were used to encapsulate recombinant human bleomycin hydrolase and transported into epithelial cells to prevent pulmonary fibrosis during BLM-based chemotherapies. The encapsulation of rhBLMH in the MHP-UiO-66 NPs protected the enzyme and enhanced cellular uptake, providing more efficient lung protection against BLMs.

Summary: This study investigates the molecular mechanism of interleukin-11 (IL-11) in silica-induced lung inflammation and fibrosis. It is found that IL-11 expression increases significantly in mouse lungs after silica exposure, mainly in lung epithelial cells and fibroblasts. Blocking IL-11 effectively reduces the expression of pro-inflammatory cytokines and fibrotic proteins induced by silica particles. The study suggests that targeting IL-11 may be a promising therapeutic approach for silica-induced lung inflammation and fibrosis.

Summary: The aim of this study is to investigate the roles of CD11b+ monocytes/macrophages in the progression of pulmonary fibrosis. The expression levels of CD11B gene and inflammatory genes in IPF patients are evaluated. CD11b cells are conditionally depleted in CD11b-DTR mice and pulmonary fibrosis is induced using bleomycin. The results show that CD11b+ monocytes/macrophages contribute to pulmonary fibrosis and represent a potential therapeutic target for IPF.

Summary: This study found that m6A methylation plays an important role in the formation of silicosis, and long-term exposure to crystalline silica can lead to silicosis accompanied by increased levels of m6A methylation. Upregulation and downregulation of certain m6A modification-related genes may contribute to aberrant m6A modification. In addition, there are genes showing differential m6A methylation peaks in silicosis, and there are 1091 genes with differential expression in silicosis.
