Summary: Multiparameter MR fingerprinting (MRF) can quantify multiple relaxation parameters of intervertebral disc (IVD) simultaneously, which may vary by age and sex. This study investigated age- and sex-related differences in the relaxation parameters of the lumbar spine's IVD using a multiparameter MRF technique. The results showed that the 3D-MRF technique has potential to characterize age-related differences in T-1, T-2, or T-1 rho of IVD in healthy subjects.

Summary: HtrA1 is an important protease involved in various biological functions and plays a key role in many human diseases. Studies have shown that HtrA1's role in degrading articular cartilage and regulating cell signaling pathways has sparked great interest in its involvement in skeletal diseases. By studying the role of HtrA1 in conditions such as osteoporosis, rheumatoid arthritis, osteoarthritis, and intervertebral disc degeneration, new approaches for prevention and treatment of these diseases can be discovered.

Summary: Bone diseases are often caused by abnormal metabolism and death of bone cells, with mitochondrial dysfunction being an important factor. Recent studies suggest that mitophagy plays a crucial regulatory role in bone diseases, indicating that regulating mitophagy levels may be a new strategy for treating bone-related diseases.

Summary: This review summarizes the application of single-cell techniques in the study of musculoskeletal disorders. It provides a review of cell isolation, experimental methods, and feasible experimental designs for single-cell studies in musculoskeletal system diseases based on tissue characteristics. Furthermore, it highlights the latest findings of single-cell studies in musculoskeletal disorders and discusses the potential application and future development directions of single-cell and spatial omics in research on musculoskeletal diseases.

Summary: Genistein, a natural isoflavone, exhibits various biological effects and has been investigated for its potential therapeutic benefits in bone and cartilage diseases. It shows estrogen-like activity in protecting against osteoporosis and osteoarthritis, and also acts as an inhibitor of tyrosine kinase involved in pathological development of these diseases. Multiple signaling pathways are involved in its regulatory activity, but the molecular mechanisms are not fully understood yet.

Summary: Degenerative musculoskeletal disorders are age-related diseases that severely affect patients' ability to work and can have serious consequences. Notch signaling pathway plays a crucial role in these disorders, retarding the progress of degeneration and having important functions in stem/progenitor cells.

Summary: The study shows that discectomy for upper lumbar disc herniations has an overall success rate of approximately 80% and has not improved with minimally invasive surgical techniques. Discectomy for herniations at L3-4 trends toward better outcomes compared with L1-2 and L2-3, but was not significant.

Summary: Musculoskeletal disorders of the cervical spine have increased in recent years. This study aimed to develop a dynamic model of the cervical spine and estimate the differences in disc loads, translations, intradiscal pressure, and muscle force/activity in a neutral posture. The results could help distinguish between normal and pathological cervical spine conditions.

Summary: RNA-binding proteins (RBPs), which consist of multiple intermediate sequences, play important roles in various biological functions by interacting with proteins, mRNA, and non-coding RNAs. This article focuses on the functions of RBPs in degenerative joint diseases (IVDD and OA) and reviews the latest advancements in understanding the regulatory mechanisms of RBPs. It provides a novel perspective on the pathophysiology, early diagnosis, and treatment of OA and IVDD.

Summary: This study characterizes alterations to human facet cartilage and subchondral bone across a spectrum of degeneration and investigates correlations between disc and facet degeneration. The findings suggest that the mechanical properties of facet cartilage and the structural properties of facet subchondral bone change with increasing degenerative grade and are correlated with aspects of disc degeneration.

Summary: Macrophage infiltration and polarization play a crucial role in lumbar intervertebral disc herniation (LDH). This study examined the association between macrophage polarization in herniated nucleus pulposus (NP) tissue of LDH patients and different clinical characteristics or symptoms. The findings suggest that macrophage polarization or macrophage-like cells contribute to the pathological features of LDH and that certain macrophage populations are associated with pain intensity during LDH. These results can potentially lead to personalized pharmacological interventions for LDH patients considering pain heterogeneity.

Summary: Ferroptosis, a form of cell death involving iron metabolism and lipid peroxidation, differs from other forms such as apoptosis, autophagy, pyroptosis, and cuproptosis. It plays a crucial role in various diseases including malignant tumors, neurodegenerative diseases, and cardiovascular diseases. Both iron overload and lipid peroxide accumulation contribute to musculoskeletal diseases, providing new opportunities for clinical diagnosis and treatment.

Summary: Ghrelin is a brain-gut peptide that can promote the release of growth hormone, potentially playing a role in bone growth and development. Recent studies have reported a correlation between ghrelin and degenerative diseases of the musculoskeletal system, suggesting that ghrelin may be a factor influencing degenerative pathologies. With the aging population, the risk of health problems caused by degenerative diseases of the musculoskeletal system increases. This article summarizes the roles of ghrelin in musculoskeletal disorders, highlighting the potential effects of ghrelin as a key target in the treatment of related bone and muscle diseases and the need for further research.

Summary: This study aims to evaluate the feasibility of using bending films to determine the lowest instrumented vertebra (LIV), and provide real-world data on outcomes. The results show that adjusting the position of the fusion vertebra based on bending films can effectively reduce the risk of postoperative spinal decompensation.
