Factoring neurotrophins into a neurite-based pathophysiological model of schizophrenia

Neurotrophins are growth factors that, through variations in concentration and changes in receptor expression, regulate the formation of axons and dendrites during development and throughout adult life. Here we review these growth factors, particularly in the context of schizophrenia, a psychiatric disorder characterized by neurodevelopmental abnormalities. We first discuss emerging information derived from physiologically relevant organotypic cultures and in vivo studies regarding the effects of neurotrophins on the neuronal structure including pruning and GABAergic neurons. We then review postmortem studies of neurotrophin levels and their receptors in brains of individuals with schizophrenia, and compare them with what is known about neurotrophin effects on neuronal structure. This comparison indicates that only some neuropathological defects encountered in patients with schizophrenia can be explained by the single action of neurotrophins on dendrites and axons. However, we propose that a number of inconsistent findings and apparently unrelated results in the schizophrenia field can be reconciled if neurons are considered structurally plastic cells capable of extending and retracting dendrites and axons throughout life. Early detection and intervention for young people with emerging bipolar disorder: What is the state of affairs?

Summary: Bipolar disorder is a disabling chronic illness with a delay in diagnosis and access to adequate care, resulting in many consequences. Early detection and intervention are essential to improve prognosis, with the identification of risk factors for BD aiming to prevent disease development, delay onset, and improve course through timely treatment. Validated tools and coordinated efforts are assets for early identification of at-risk individuals, with targeted primary prevention interventions offered by frontline mental health professionals and early secondary prevention provided by general psychiatric or general medicine services.

Summary: A genome-wide association study identified significant genetic loci associated with suicide attempts, particularly one locus on chromosome 7 which is implicated in risk-taking behavior, smoking, and insomnia. The study also revealed strong genetic correlations between suicide attempts and psychiatric disorders, as well as other nonpsychiatric traits like smoking, pain, and lower socioeconomic status. Conditioning on psychiatric disorders reduced the genetic correlations between suicide attempts and psychiatric disorders, indicating a shared biology with nonpsychiatric risk factors.

Summary: Dysfunction in the H-PFC circuit is a critical factor in schizophrenia, and a new drug called EGIS 11150 has been found to induce theta rhythm, enhance H-PFC coherence, and restore long-term potentiation in this circuit. This suggests that phenotypical screening on H-PFC connectivity can uncover novel antipsychotics.

Summary: Current treatments for trauma-related disorders are ineffective for many patients. This study modeled individual differences in post-therapy fear relapse using an ethologically relevant trauma recovery paradigm. The results suggest that post-trauma behavioral phenotypes and gene expression patterns are associated with fear relapse susceptibility, which may be important for future treatment development.

Different dimensions of caregiving burden have varying impacts on depressive symptoms, with negative emotion/consequences, (lack of) positive personal experience, and stigma/effects on family being the most connected to depression.

Summary: Schizophrenia is influenced by both genetic and environmental factors, with adolescence being a critical period for symptom manifestation. This study focused on the interaction between DNA methylation and mRNA expression, using the MAM animal model to study neurodevelopmental disruptions and behavioral deficits. The results showed reduced expression levels of Drd2 and high alterations of methylation rates in co-factor genes under MAM treatment during pre-adolescence.

Summary: Individuals with clinical high risk for psychosis show deficits in verbal memory and verbal fluency, which are related to longer-term outcomes of the disorder. These cognitive impairments are present even in those not taking antipsychotic medication. Furthermore, impaired immediate verbal recall predicts functional disability and non-remission from the clinical high risk state.
