Chromene stability: The most potent estrogenic compounds in White Kwao Krua (Pueraria candollei var mirifica) crude extract

Pueraria candollei var. mirifica (PM) is a phytoestrogen-rich herb with rejuvenating qualities. Stability of chromenes, comprising deoxymiroestrol, miroestrol and isomiroestrol, in crude extract of PM was investigated. The effects of acid-base condition and temperature on stability of the chromene standard solution and the PM crude extract were analysed using gradient HPLC. The degradation of the chromenes in the extract was sensitive to high temperatures and strongly acidic or alkaline solutions. Specifically, deoxymiroestrol, which exhibited the highest estrogenic activity, can be easy degraded to miroestrol or isomiroestrol. However, chromene standard solution was stable in ethanol after storage for 360 days at 25 degrees C. The crude extract of PM in the solid form was stable when stored at -20 or 4 degrees C for 360 days. Therefore, the extract could be stored in the solid form at low temperatures. This method provides a system to simultaneously identify chromenes in crude extract of PM. (C) 2015 Elsevier Ltd. All rights reserved.

The tissue cultures produced compounds that exhibited anti-inflammatory effects, suggesting they could be a valuable and sustainable source of active ingredients.

Summary: This study presents the design of hydrophobic deep eutectic solvents (HDESs) and HDES-based microemulsions for the extraction of curcuminoids and ar-turmerone from Curcuma longa L. The results show that HDES and its microemulsions are more effective and environmentally friendly than conventional organic solvents, and they are also compatible with food and pharmaceutical formulations.

Comparative stability and analytical performance of anti-miroestrol recombinant antibody in different cassettes

Summary: This study developed and validated an ICA with a monoclonal antibody exhibiting similar reactivity against Mi and Dmi. The ICA showed a LOD at 1 and 16 mu g/mL for Mi and Dmi, respectively. The cross-reactivity of the ICA with Dmi was lower than that observed with the icELISA, and the cross-reactivity of ICA against other compounds of the PM was correlated with those of icELISA.

Construction of a monoclonal antibody against glabridin (2G4) and development of an enzyme-linked immunosorbent assay

Summary: This study aimed to develop an enzyme-linked immunosorbent assay (ELISA) using a glabridin-specific antibody. The glabridin-bovine serum albumin conjugates were injected into mice to produce hybridomas, and a highly specific antibody against glabridin was produced. The developed ELISA method showed high sensitivity and specificity for quantifying glabridin in plant materials and human serum samples.

An eco-friendly method using deep eutectic solvents immobilized in a microcrystal cellulose-polyvinyl alcohol sponge for parabens analysis in food samples

Summary: An eco-friendly and reusable DESs-MCC/PVA sponge was synthesized and showed good efficiency and reproducibility in the extraction of parabens in milk, beverages, and tea samples. The sponge retained > 80% extraction efficiency and could be reused up to five times. This method offers a simple and environmentally friendly approach for the extraction of trace organic substances.

Summary: Micellar liquid chromatography (MLC) was used to analyze low concentration canthin-6-one alkaloids in extracts of Eurycoma longifolia Jack (EL) and Eurycoma harmandiana Pierre (EH). The MLC method showed comparable sensitivity, precision, accuracy, and acetonitrile consumption to the conventional high-performance liquid chromatography (HPLC) method. However, the HPLC method exhibited better performance for samples with low analyte concentrations and varying sample matrices, especially EL and EH samples. Overall, the MLC analysis of canthin-6-one alkaloids was limited to trace analytes due to interference by the matrix.

Impact of elicitors and precursor on quassinoids and canthin-6-ones production in adventitious root culture of Eurycoma harmandiana Pierre and improvement of their anti-inflammatory activity

Summary: The study investigated the growth kinetics of adventitious root culture of E. harmandiana and the effect of various elicitors and precursors on the contents of bioactive compounds. It was found that the elicitors and precursor significantly increased the accumulation of quassinoids and canthin-6-ones, leading to stronger anti-inflammatory effects.
