IL-13 Induces YY1 through the AKT Pathway in Lung Fibroblasts

A key feature of lung fibrosis is the accumulation of myofibroblasts. Interleukin 13 (IL-13) is a pro-fibrotic mediator that directly and indirectly influences the activation of myofibroblasts. Transforming growth factor beta (TGF-beta) promotes the differentiation of fibroblasts into myofibroblasts, and can be regulated by IL-13. However, IL-13's downstream signaling pathways are not completely understood. We previously reported that the transcription factor Yin Yang 1 (YY1) is upregulated in fibroblasts treated with TGF-beta and in the lungs of mice and patients with pulmonary fibrosis. Moreover, YY1 directly regulates collagen and alpha smooth muscle actin (alpha-SMA) expression in fibroblasts. However, it is not known if IL-13 regulates fibroblast activation through YY1 expression. We hypothesize that IL-13 upregulates YY1 expression through regulation of AKT activation, leading to fibroblast activation. In this study we found that YY1 was upregulated by IL-13 in lung fibroblasts in a dose- and time-dependent manner, resulting in increased alpha-SMA. Conversely, knockdown of YY1 blocked IL-13-induced alpha-SMA expression in fibroblasts. Furthermore, AKT phosphorylation was increased in fibroblasts treated with IL-13, and AKT overexpression upregulated YY1, whereas blockade of AKT phosphorylation suppressed the induction of YY1 by IL-13 in vitro. In vivo YY1 was upregulated in fibrotic lungs from CC10-IL-13 transgenic mice compared to that from wild-type littermates, which was associated with increased AKT phosphorylation. Taken together, these findings demonstrate that IL-13 is a potent stimulator and activator of fibroblasts, at least in part, through AKT-mediated YY1 activation.

Divergent Regulation of Alveolar Type 2 Cell and Fibroblast Apoptosis by Plasminogen Activator Inhibitor 1 in Lung Fibrosis

Summary: The increased sensitivity to apoptosis of alveolar type 2 cells (ATII) and the increased resistance to apoptosis of (myo)fibroblasts, known as the apoptosis paradox, could be attributed to the regulation of plasminogen activator inhibitor 1 (PAI-1), which is up-regulated in both cell types in idiopathic pulmonary fibrosis (IPF). The findings suggest that the age-related changes in PAI-1 levels may underlie the apoptosis sensitivity and resistance in ATII and fibroblast cells in IPF lungs. The genetic deletion of PAI-1 in mice led to alterations in p53, p21, and Bax protein expression, impacting the apoptosis response in these cells and ultimately affecting lung fibrosis development.

Citrullinated vimentin mediates development and progression of lung fibrosis

Summary: Environmental exposures of cadmium and carbon black were found to increase in lung tissue of idiopathic pulmonary fibrosis (IPF) subjects, leading to higher levels of citrullinated vimentin (Cit-Vim). Cit-Vim was associated with higher expression in IPF patients, especially smokers, and induced fibroblast invasion and fibrosis related gene expression. These findings suggest a potential role for Cit-Vim as a damage-associated molecular pattern molecule in lung fibrosis caused by environmental exposures.

PAI-1 Regulation of p53 Expression and Senescence in Type II Alveolar Epithelial Cells

Summary: Cellular senescence, especially the senescence of alveolar epithelial type II (ATII) cells, plays a crucial role in aging and aging-related diseases such as idiopathic pulmonary fibrosis (IPF). Plasminogen activator inhibitor 1 (PAI-1), a serine protease inhibitor, was found to bind to proteasome components and inhibit proteasome activity, leading to ATII cell senescence by preventing p53 degradation. This study provides insights into the molecular mechanisms underlying ATII cell senescence and its potential implications for the pathogenesis of lung fibrosis and other age-related diseases.
