Enterovirus 71 Induces Mitochondrial Reactive Oxygen Species Generation That is Required for Efficient Replication

Redox homeostasis is an important host factor determining the outcome of infectious disease. Enterovirus 71 (EV71) infection has become an important endemic disease in Southeast Asia and China. We have previously shown that oxidative stress promotes viral replication, and progeny virus induces oxidative stress in host cells. The detailed mechanism for reactive oxygen species (ROS) generation in infected cells remains elusive. In the current study, we demonstrate that mitochondria were a major ROS source in EV71-infected cells. Mitochondria in productively infected cells underwent morphologic changes and exhibited functional anomalies, such as a decrease in mitochondrial electrochemical potential Delta Psi(m) and an increase in oligomycin-insensitive oxygen consumption. Respiratory control ratio of mitochondria from infected cells was significantly lower than that of normal cells. The total adenine nucleotide pool and ATP content of EV71-infected cells significantly diminished. However, there appeared to be a compensatory increase in mitochondrial mass. Treatment with mito-TEMPO reduced eIF2 alpha phosphorylation and viral replication, suggesting that mitochondrial ROS act to promote viral replication. It is plausible that EV71 infection induces mitochondrial ROS generation, which is essential to viral replication, at the sacrifice of efficient energy production, and that infected cells up-regulate biogenesis of mitochondria to compensate for their functional defect.

Derangements and Reversibility of Energy Metabolism in Failing Hearts Resulting from Volume Overload: Transcriptomics and Metabolomics Analyses

Summary: This study investigates the changes and reversibility of cardiac energetics in a rat model of cardiac volume overload (VO) using transcriptomics and metabolomics analyses. The findings suggest that glycolysis is downregulated and redirected towards the hexosamine biosynthetic pathway (HBP) and O-linked-N-acetylglucosaminylation in the compensated hypertrophy (CH) phase, and further suppressed in the progression to heart failure (HF). Derangements in fatty acid oxidation become prominent only in HF. Correction of VO can reverse the differential expression of genes involved in glycolysis, HBP, and fatty acid oxidation in CH but not in HF.

Kynurenine Pathway of Tryptophan Metabolism Is Associated with Hospital Mortality in Patients with Acute Respiratory Distress Syndrome: A Prospective Cohort Study

Summary: Acute respiratory distress syndrome (ARDS) is characterized by dysregulated immune-inflammatory responses and high mortality. This study found that the activation of the kynurenine pathway, a metabolic pathway, is associated with mortality in ARDS patients. Plasma kynurenine levels and the kynurenine/tryptophan ratio were higher in non-survivors compared to survivors. Urine metabolic profiling also revealed higher tryptophan degradation in non-survivors. These findings suggest that the kynurenine pathway could serve as a prognostic marker and a target for therapeutic interventions in ARDS.

Malonyl-CoA Accumulation as a Compensatory Cytoprotective Mechanism in Cardiac Cells in Response to 7-Ketocholesterol-Induced Growth Retardation

Summary: 7-Ketocholesterol (7KCh), the major oxidized product of cholesterol, induces cellular oxidative damage. This study investigated the physiological responses of cardiomyocytes to 7KCh. Treatment with 7KCh inhibited the growth of cardiac cells and their mitochondrial oxygen consumption, while also resulting in compensatory increase in mitochondrial mass and adaptive metabolic remodeling. The accumulation of malonyl-CoA was found to play a role in the growth inhibitory effect of 7KCh.
