The review also highlights the prospects and challenges associated with nanodelivery platforms in addressing cisplatin resistance in HNSCC.

Summary: This study found that tumor necrosis factor alpha-induced protein 2 (TNFAIP2) plays a crucial role in cisplatin-based chemotherapy resistance in head and neck squamous cell carcinoma (HNSCC). TNFAIP2 protects cancer cells from cisplatin-induced apoptosis by inhibiting ROS-mediated c-JUN N-terminal kinase (JNK) phosphorylation. Moreover, TNFAIP2 competes with KEAP1 to prevent the degradation of NRF2, resulting in NRF2 accumulation and cisplatin resistance.

The results showed that VE-822 inhibited cell proliferation, colony formation, and migration abilities, and improved the chemotherapeutic effect of cisplatin by inhibiting p-ATR expression and blocking DNA damage repair pathway. Conclusion: VE-822 can enhance the therapeutic effect of cisplatin on head and neck squamous cell carcinoma by inhibiting p-ATR expression.
