A Magnetic Nanoparticles-Based Combination Detection of COX-2 and BCL-2 Polymorphisms Associated with Gastric Cancer Susceptibility

In China, gastric cancer is still the main cause of death in patients with malignant tumors. Cyclooxygenase-2 (COX-2) and B-cell lymphoma-2 (BCL-2) overexpression has recently been reported as a potential tumor initiator or promoter implicated in gastric carcinogenesis. In this study, we genotyped the COX-2 -765G > C and BCL-2 -938C > A polymorphisms in 118 gastric cancer patients and 120 healthy controls, using a method based on gamma-Fe2O3 magnetic nanoparticles (MNPs) and dual-color fluorescence hybridization. The associations between COX-2 -765G > C and BCL-2 -938C > A polymorphisms and risk of gastric cancer have been investigated. Samples with COX-2 gene -765GC and -765CC genotypes were significantly higher than that with COX-2 gene 765GG genotype in gastric cancer group (OR = 1.808, 95%Cl: 1.050-3.113, P = 0.033). Therefore, the carriers of C allele of COX-2 -765G/C polymorphism are more prone to gastric cancer in comparison with non-carriers. The gastric cancer individuals carrying BCL-2 938CC genotype have a 3.185-fold risk of gastric cancer than those carrying the AA genotype. In comparison with COX-2 -765GG and BCL-2 -938AA, risk of gastric cancer in COX-2 -765CC and BCL-2 938CC, COX-2 -765GC and BCL-2 938CC is significantly higher (OR= 10, 95% Cl: 1.280-78.117, P = 0.028; OR = 4.667, 95% CI: 1.137-19.154, P = 0.033). The genetic polymorphism of COX-2 -765G > C and BCL-2 -938C > A may jointly affect the onset of gastric cancer.
