Summary: Photodynamic therapy (PDT) is an emerging clinical treatment that is expected to become an important adjuvant strategy for immunotherapeutic cancer treatment. However, the immune response of PDT is not lasting and eventually turns into immunosuppression, which limits its further development. This study demonstrates that high-light-dose PDT induces early immune activation followed by late immunosuppression mediated by upregulated TGF-beta 1. The mechanism of PDT-induced TGF-beta 1 accumulation and immunosuppression is elucidated, and a TGF-beta blockade treatment strategy is presented as a promising approach to reverse PDT-associated immunosuppression.

Summary: The aim of this study was to investigate the effects and mechanisms of Qingda granule (QDG) treatment on hypertensive vascular remodeling in vivo and in vitro. The results showed that QDG significantly attenuated arterial wall thickening and collagen expression in hypertensive rats. QDG also inhibited cell migration and collagen expression in stimulated fibroblasts. These findings suggest that QDG may have therapeutic potential for hypertensive vascular remodeling.

Summary: Angiotensin II upregulates TGF-beta 1 and ET-1, which act as profibrotic mediators. The signal transduction of ATR for upregulation of TGF-beta 1 and ET-1, and their effectors in myofibroblast differentiation, is not fully understood. In this study, the ATR networking with TGF-beta 1 and ET-1 was investigated, and the signal transduction of these mediators was identified. The findings suggest that TGF-beta 1 and ET-1 are major effectors of the ATR cascade and negative regulation of their signaling represents a targeted therapeutic strategy for cardiac fibrosis prevention and restoration.

Summary: Cigarette smoke induces the deterioration of airway smooth muscle cells (ASMCs) in chronic obstructive pulmonary disease (COPD), leading to tissue loss and structural remodeling of the bronchi. The study found that COPD ASMCs showed decreased expression of collagen type-I and alpha-smooth muscle actin (alpha-SMA), but increased levels of fibronectin. Cigarette smoke extract (CSE) further downregulated collagen type-I and alpha-SMA expression, but upregulated fibronectin. CSE-induced mitophagy and lysosome activity were mediated by ERK1/2 phosphorylation.
