Summary: The iron complexes were found to inhibit cell proliferation, reduce metabolic activity, and increase mitochondrial reactive oxygen species. The mode of action involved ferroptosis along with lipid peroxidation and diminished mitochondrial membrane potential. All compounds showed similar activity except for compound 4, which was slightly less active. Electrochemical analyses showed fast attachment of the solvent dimethyl sulfoxide and release of the axial ligand A for all compounds.

The production of HO· was quantified by a chemiluminescence method, showing that [Fe-IV(O)(tpena)](+) is produced in much larger yields than HO·. Additionally, the study indicates that a reactive high-valent iron-oxo species and HO· can be generated under mild, aqueous ambient conditions, which represents a significant contribution to biomimetic nonheme chemistry in water.
