Summary: This study comprehensively analyzed the epigenetic regulation in rheumatoid arthritis (RA) using differential analysis and cross-analysis. The results identified differentially methylated sites, differentially expressed genes, and differentially expressed miRNAs associated with RA. A comprehensive epigenetic regulatory network was constructed, and core regulatory genes were identified. This study provides a new direction for future research on the epigenetic mechanisms of RA.

CCL2, a chemokine and activator of monocytes, macrophages, and neutrophils, plays a key role in amplifying and accelerating the inflammatory cascade and is closely related to chronic non-controllable inflammation. Understanding the regulatory mechanisms of CCL2 can provide potential targets for the treatment of inflammatory diseases.

Summary: The study showed that MSLN-TTC has immunostimulatory effects both in vitro and in vivo, demonstrating significant antitumor activity when used alone and enhanced efficacy when combined with an inhibitor of the immune checkpoint programmed death receptor ligand 1 (PD-L1). The activation of immune cells, including DCs and CD8+ T cells, contributes to the antitumor efficacy of MSLN-TTC.

Summary: This study investigates the combinatorial mechanisms of genome activation and shows that zebrafish genome activators Pou5f3 and Nanog exhibit different roles in the activation process. They act as pioneer-like transcription factors on synergistic enhancers, promoting histone acetylation and transcription activation. However, on antagonistic enhancers, one of the factors acts as a non-pioneer-like transcription factor, competing with the activator and blocking its effects. This activator-blocker mechanism restricts widespread transcriptional activation and prevents premature expression of late developmental genes in early embryos.

Summary: This study investigated the immunostimulatory effects of collagen and chitosan preparations extracted from fishery discards on human peripheral blood mononuclear cells (PBMCs). The results showed that these preparations did not exhibit cytotoxic effects, but they promoted the release of cytokines and increased the production of cellular ROS. They also led to the activation of T CD8(+) and CD4(+) lymphocytes, as well as an increase in cytosolic calcium concentration and mitochondrial membrane potential.

Summary: The field of RNA modification, known as epitranscriptomics, is attracting more attention from scientists. Over 160 chemical modifications have been found in RNA molecules, but their functions are still unclear. This review focuses on the role of N-6,2'-O-dimethyladenosine (m(6)A(m)) in gene expression regulation. While the role of m(6)A(m) in splicing regulation has been supported by studies, conflicting data exist regarding its role in mRNA stability and translation. Nonetheless, promising results have been obtained by modulating regulators in cancer cells. Further investigation on m(6)A(m) is expected to yield significant findings.

Summary: Accurate and precise regulation of gene expression in eukaryotes involves the coordinated control of transcription, mRNA stability, and translation. Recent progress has been made in understanding the role of sequence elements in the 3' untranslated region in mRNA degradation regulation. The recruitment of the Ccr4-Not complex to target mRNA is a critical step in mRNA decay regulation. This article reviews the role of the human Ccr4-Not complex in cytoplasmic deadenylation of mRNA, focusing on its recruitment mechanisms and general mechanisms involving the poly(A) tail.
