Hygromycin promotes somatic embryogenesis in spinach

Hygromycin (hyg) at low doses (0.5-1.0 mg l(-1)) promoted somatic embryogenesis from apical sections of spinach lateral roots. The highest promoting effect on both the frequency of regeneration and the mean number of somatic embryos (SE) per explant was achieved at 0.5 mg l(-1) hyg. With increasing the concentration of hyg to 1 mg l(-1), the regeneration frequency decreased, while the mean SE number remained significantly higher than in control (hyg-free medium). Complete inhibition of SE regeneration started at 7.5 mg l(-1) hyg. Moreover, hyg efficiently promoted the process of secondary somatic embryogenesis. Compared to control, a 2.75-fold increase in the secondary somatic embryo (SSE) mean number was obtained at 0.5 mg l(-1) hyg, and the increment was still discernible at 1.0 and 2.5 mg l(-1) hyg. Both primary SE and SSE explants became completely necrotic at 12.5 mg l(-1) hyg. Since attempts with direct selection at 20 mg l(-1) hyg proved unsuccessful, the results obtained in this study suggest that a stepwise selection procedure is suitable, starting with selection at 0.5 mg l(-1) hyg, to exploit the promoting effect of low hyg doses on SE regeneration from transformed cells, then gradually increasing the hyg concentration to 20 mg l(-1) for final selection. Complete SE and SSE explant mortality at hyg above 12.5 mg l(-1) guarantees a low possibility of escape during the selection process. This study will be useful for increasing the efficiency of transgenic plant regeneration following genetic transformation in spinach.

Transcriptome Profiling of the Potato Exposed to French Marigold Essential Oil with a Special Emphasis on Leaf Starch Metabolism and Defense against Colorado Potato Beetle

Summary: A microarray transcriptome analysis was conducted to study the effects of French Marigold essential oil on potato metabolism, revealing that exposure to the oil suppressed genes related to photosynthesis and altered leaf starch metabolism. Additionally, the exposure led to an increase in sucrose accumulation in potato plants compared to glucose and fructose. The changes in transcriptional levels and sugar metabolism caused by the essential oil also resulted in enhanced feeding behavior and development of Colorado potato beetle larvae.

Impact of Storage Temperature on Pollen Viability and Germinability of Four Serbian Autochthon Apple Cultivars

Summary: This study found that the viability and germination rate of fresh pollen varied under different storage temperatures, with Budimka cultivar showing the highest viability and germination rate. Storage at 4 degrees Celsius extended the pollen viability and germinability, while -20 or -80 degrees Celsius significantly prolonged the pollen longevity for all cultivars. After 6 months, pollen stored at -20 and -80 degrees Celsius had much higher germination rates compared to pollen stored at room temperature for the same period.

Germination and the Initial Seedling Growth of Lettuce, Celeriac and Wheat Cultivars after Micronutrient and a Biological Application Pre-Sowing Seed Treatment

Summary: Seed treatments with zinc, boron, biostimulant Coveron and MIX have shown positive effects on seed germination and seedling growth of various lettuce, celeriac, and wheat cultivars, while also inhibiting the growth of pathogens. Different cultivars showed varying responses to the seed treatments, with significant interactions observed between cultivars and treatments. The relationship between seed germination and seedling growth was found to be significant, while the relationship between seed germination and pathogens was negative for all cultivars.

Summary: Species of the genus Aesculus, such as horse chestnut, have medicinal uses and can be used for the treatment of venous insufficiency. Various in vitro propagation methods have been established for mass production of embryos and shoots, including somatic embryogenesis and shoot organogenesis. Additionally, an efficient method for cryopreservation of embryogenic tissue has been developed to ensure constant availability of the plant material.

In Vitro Shoot Multiplication and Regeneration of the Recalcitrant Rocket (Eruca sativa Mill.) Variety Domaca Rukola

Summary: In this study, an efficient tissue culture system for improving the rocket cultivar Domaca rukola was established. Optimal shoot multiplication and organogenesis protocols were determined through experiments with different hormone concentrations and combinations. In addition, in vitro-produced rocket shoots were found to be a potential source of the antioxidant flavonoid kaempferol.

Phytohormone Profiling of Potato (Solanum tuberosum L.) Exposed to French Marigold (Tagetes patula L.) Essential Oil

Summary: The study investigated the comprehensive phytohormone profile of potato plants exposed to French marigold essential oil (EO) for different time periods. The results showed altered phytohormone responses of EO-exposed potato plants, with stress-related phytohormones being the most affected. The expression levels of genes involved in biosynthesis and catabolism of stress-related phytohormones indicated that the jasmonic acid biosynthetic pathway was the most affected.

The pyruvate dehydrogenase complex regulates mitophagic trafficking and protein phosphorylation

Summary: The mitophagic degradation of mitochondrial matrix proteins in Saccharomyces cerevisiae is selectively regulated by the matrix kinases Pkp1 and Pkp2, which are in turn regulated by the phosphatase Aup1/Ptc6. These proteins also regulate the phosphorylation status and catalytic activity of the pyruvate dehydrogenase complex, which is critical for mitochondrial metabolism. The pyruvate dehydrogenase complex has been found to have a novel function in regulating mitophagic selectivity independent of its enzymatic activity.

Somatic Embryogenesis in Spinach-A Review

Summary: A spinach-supplemented diet offers various health benefits, but caution is necessary due to the potential health issues from high levels of oxalic acid and nitrate. Breeding can increase the levels of vitamins and phytochemicals, but conventional breeding is constrained by complex sex determination. By utilizing tissue culture techniques and biotechnological methods, limitations in spinach somatic embryogenesis can be overcome, providing a pathway for further research and improvement in the efficiency of this process.

Overcoming Low Germination and Low Quality of Flax Seeds (Linum usitatissimum L.) in Unfavorable Storage Using Static Magnetic Fields

Summary: This study demonstrates that exposing flax seeds to static magnetic fields can improve their germination and quality. Specifically, a 350 mT magnetic field treatment for 100 and 120 minutes achieved the best germination results and showed significant improvements in several biological indicators.
