Summary: The study aims to investigate the role of GRK4 in the pathogenesis and progression of myocardial infarction (MI). Results showed that GRK4 expression was increased in the heart after MI, and overexpression of GRK4 aggravated cardiac infarction and dysfunction while specific gene knockout ameliorated these effects. GRK4 inhibited autophagy and promoted cardiomyocyte apoptosis, effects mediated by HDAC4 phosphorylation and a decrease in beclin-1 expression, leading to greater impairment of cardiac function in MI patients carrying the GRK4 A486V variant.

Summary: This study investigates the effects of S100A9 blockade on the biological processes during recovery after myocardial infarction (MI). The results suggest that blockade of S100A9 regulates the expression of proteins involved in leukocyte cell-cell adhesion, muscle cell apoptosis, cardiac muscle hypertrophy, and other biological processes. It prevents compensatory hypertrophy and reduces post-ischemic stress in the heart.

Summary: The interplay between apoptosis and senescence is crucial in cancer development, and targeting the Bcl-2 family proteins could be a promising strategy for cancer therapy. Therapy-induced senescence (TIS), induced by chemotherapeutic agents, has been controversial in its effect on therapeutic outcome. Clearance of senescent cells and overcoming their pro-survival mechanisms are important challenges in cancer treatment. This review article discusses recent literature on the role of the Bcl-2 family proteins in apoptosis and senescence, and the progress and limitations in targeting them for cancer therapy.
