Genistein aglycone effect on bone loss is not enhanced by supplemental calcium and vitamin D3: A dose ranging experimental study

Genistein aglycone (GEN) has a favorable effect on bone loss. We investigated the effects of GEN alone or in combination with supplemental calcium and vitamin D-3 in an animal model of bone loss to evaluate if there was additional benefit. Ovariectomized (OVX) and SHAM-OVX rats were used. OVX were divided into 12 groups and randomized to receive: GEN at 27, 54, 200, 500 or 1000 mg (human equivalent dose (HED)/day/ip injection alone or with calcium carbonate (Ca)(360 mg/kg/day/gavages)and vitamin D-3 (D-3) (50 IU/kg/day/gavages) or Ca/D-3 without GEN or untreated for 6 weeks. SHAM-OVX were randomized into 7 groups and treated with: Ca and D-3 alone or in combination with GEN (same doses as OVX), or left untreated. Bone mineral density (BMD), bone-alkaline phosphatase (b-ALP), collagen C-telopeptides (CTX), osteoprotegerin (OPG) and soluble receptor activator of NF kappa B ligand (sRANKL) were assessed. Femurs were excised and tested for breaking strength and histology. Uterine weight was analyzed to assess GEN's estrogenic effects on the SHAM-OVX. The most effective dose of GEN, independent of Ca/D-3 supplementation, was 54 mg/day. Higher closes yielded no further improvement in bone biomarkers. histology or strength. Only 1000 mg/day HED of genistein produced statistically significant changes in uterine weight of the SHAM-OVX. This study suggests that 54 mg/day of GEN is the threshold dose for efficacy. In addition, supplemental calcium and vitamin D-3, beyond normal dietary intake do not enhance the effects of genistein on improving measures of bone loss. This observation has implications regarding the use of calcium and vitamin D-3 supplementation. (C) 2011 Elsevier GmbH. All rights reserved.

Summary: In multiple myeloma, mesenchymal stromal cells (MSCs) promote tumor growth and provide metabolic support to plasma cells (PCs) through mitochondrial transfer. Myeloma MSCs rely less on mitochondrial metabolism compared to healthy MSCs and are more prone to delivering mitochondria to myeloma cells. Additionally, PCs induce the expression of connexin 43 (CX43) in MSCs, leading to the activation of CXCL12 and the stimulation of its receptor CXCR4 on myeloma cells, promoting mitochondrial transfer. These findings suggest that the CXCL12/CXCR4 axis mediates intercellular communication in the myeloma niche.

Summary: About 30% of patients with papillary thyroid cancer (PTC) experience persistent or recurrent disease within 10 years after initial treatment. However, the measurement of serum thyroglobulin (Tg) is unreliable in the presence of anti-thyroglobulin antibodies (TgAb). This pilot study investigated the potential of circulating miRNA as valuable biomarkers for the early detection of persistent disease in TgAb-positive PTC patients. The findings indicate that serum miRNA kinetics may provide additional and independent information for the early detection of persistent disease in PTC patients with uninformative Tg results.

Nutraceuticals as Alternative Approach against Cadmium-Induced Kidney Damage: A Narrative Review

Summary: Cadmium (Cd) poses a risk to public health due to its non-biodegradability and long half-life in the body. Cd mainly targets the kidneys and accumulates there. Studies have shown that Cd exposure can lead to bone fragility, not only through direct toxic effects on bone mineralization but also due to renal failure. Further research has revealed the molecular pathways through which Cd induces serious kidney damage, including lipid peroxidation, inflammation, programmed cell death, and hormonal imbalances. Chronic kidney disease (CKD) is associated with dysbiosis, and recent studies have highlighted the altered composition and functions of gut microbiota in CKD. Considering the strong connection between diet and CKD management, nutraceuticals found in Mediterranean diet can be a safe therapeutic strategy for Cd-induced kidney damage and contribute to the prevention and treatment of CKD.

Summary: The present study evaluated the effects of beta-Caryophyllene (BCP) on Cd-induced neurotoxicity. The results showed that BCP treatment reduced ROS production, increased Nrf2 expression, and protected cells from death. BCP also regulated the apoptosis pathway and reduced the increase of pro-inflammatory cytokines.

Glutathione: Lights and Shadows in Cancer Patients

Summary: In cases of cellular injury, excessive production of reactive oxygen species (ROS) can lead to various conditions, including cancerogenesis. Glutathione (GSH), the most abundant thiol-containing antioxidant, is crucial for restoring redox homeostasis. The conjugation of GSH with xenobiotics, such as anti-cancer drugs, can either reduce their harmful effects or enhance their toxicity. Further studies are needed to better understand the relationship between GSH and cancer. The use of GSH should only be considered under medical supervision, taking into account the appropriate timing and setting, as self-prescribed GSH lacks strong scientific evidence for its efficacy in reducing toxicity.

Summary: Multiple myeloma (MM) is a difficult-to-treat malignant disease characterized by the proliferation of plasma cells in the bone marrow. This review analyzes the presence of neoantigens in MM patients and evaluates their potential as prognostic factors or therapeutic targets. The use of neoantigen treatment strategies and bispecific, trispecific, and conjugated antibodies for MM treatment are discussed. The utilization of CAR-T in relapsed and refractory patients is also examined.

Biglycan (BGN) plays a key role in matrix assembly and the control of cardiac fibroblast phenotype. Adenosine receptors (ARs), particularly A(2AR), may stimulate fibrotic damage through collagen production, and A(2AR) modulation could be an effective therapeutic option to reduce fibrotic processes in heart remodeling.
