Summary: Secondary poor graft function (sPGF) after haploidentical hematopoietic stem cell transplantation (HSCT) in patients with acquired aplastic anemia (AA) has a cumulative incidence of 4.62% and is associated with lower 2-year overall survival. Later neutrophil engraftment and a history of refractory cytomegalovirus viremia are risk factors for sPGF.

Summary: This study evaluated the efficacy of T-cell replete HLA haploidentical hematopoietic stem cell transplantation (TCR-haplo-HSCT) in pediatric refractory/relapsed B-cell precursor acute lymphoblastic leukemia. The results suggest that this transplantation approach has the potential to improve outcomes in some patients with RR-BCP-ALL, but it also carries certain risks.

Summary: The development of Disease Risk Comorbidity Index (DRCI) based on Disease Risk Index (DRI) and Hematopoietic Cell Transplantation-Specific Comorbidity Index (HCT-CI) allows for effective risk stratification of patients receiving haploidentical hematopoietic stem cell transplantation. The DRCI accurately predicts disease-free survival, particularly in disease-specific subgroups such as acute leukemia patients. This index also shows predictive value for relapse, non-relapse mortality, overall survival, and can aid in identifying patients who will benefit most from the transplantation procedure.

Summary: This study examined the influence of graft composition in haploidentical hematopoietic stem cell transplantation. The results showed that using G-CSF-primed bone marrow and peripheral blood stem cells had comparable outcomes to using only G-CSF-mobilized peripheral blood stem cells. Furthermore, patients with a certain range of nucleated cell transfusion had lower transplant-related mortality and slightly improved survival rates. These findings suggest that adjusting the composition and ratio of graft cells can enhance the efficacy of haploidentical hematopoietic stem cell transplantation.

Summary: Allogeneic stem cell transplantation is an effective therapy for hematopoietic malignancies, relying on donor T cells and NK cells for the graft-versus-leukemia effect. However, graft-versus-host disease remains a major complication, highlighting the importance of understanding alloantigen presentation to separate beneficial effects from detrimental ones. Mouse models and tools play a key role in quantifying alloantigen presentation post-alloSCT.

Summary: Unstable hemoglobinopathies are rare diseases that disrupt hemoglobin stability, causing chronic hemolysis and anemia. Hematopoietic stem cell transplantation has been reported as a curative approach for transfusion-dependent patients with unstable hemoglobinopathies. This case report describes the successful haploidentical HSCT treatment of Hb Bristol-Alesha variant in a 6-year-old boy with severe anemia. Two years post-transplantation, the patient achieved near normal hemoglobin levels without signs of hemolysis, confirming full engraftment with normal erythropoiesis through DNA analysis.

Summary: Post-transplantation cyclophosphamide (PTCy) is a drug used to treat graft-versus-host disease (GVHD) following hematopoietic cell transplantation (HCT). It works by impairing and suppressing antigen-specific T cells. In this study, reduced proliferation of CD4(+) T cells at day +7 and preferential recovery of CD4(+)CD25(+)Foxp3(+) regulatory T cells (T-regs) at day +21 were identified as potential biomarkers associated with optimal PTCy dosing and timing.

Summary: A new method called MASE-CE has been developed for genotyping analysis in patients with colorectal cancer. It can simultaneously detect KRAS/NRAS/BRAF gene mutations and microsatellite instability (MSI) status. The method showed high consistency with NGS and PCR-CE, and had superior sensitivity in mutation and MSI detection. This time-saving and specimen-saving method can provide timely clinical decision support.

Summary: Optical anisotropy in SnSe is utilized to design broadband and incident-angle-modulation near-infrared polarizers, with high modulation efficiency and no energy consumption. The broadband polarizer has a bandwidth of 324 nm and an average extinction ratio above 23 dB, while the incident-angle-modulation polarizer dynamically and reversibly modulates its working wavelength with a maximum extinction ratio of 71 dB. The proposed design opens up possibilities for mass-production and integration of polarizing optical components based on low-symmetry materials.

Summary: Single-cell RNA sequencing revealed that a high baseline interferon gamma (IFN-gamma) signature in CD8(+) T cells predicts better responses to neoadjuvant immunotherapy plus chemotherapy in gastric cancer (GC). The therapy also leads to a significant decrease in the IFN-gamma signature in multiple cell types and suppresses the exhausted signature of CD8(+) T cells during treatment.

Summary: The article introduces a new method for estimating blood pressure based on the iPPG pulse signals, which captures the tiny change of skin color caused by a heartbeat using consumer-level cameras. It incorporates cardiovascular characteristics such as heart rate, stroke volume, the elasticity of vessel walls, and peripheral vascular resistance. Bayesian neural network models are constructed for the estimation of systolic and diastolic blood pressure, and the machine learning uncertainty is evaluated. The uncalibrated method has been tested on 220 patients with a history of cardiovascular diseases, demonstrating its reliability with low estimation errors.

Summary: The ternary direct-current TENG (T-DC-TENG) is a promising new technology that disrupts the traditional energy production model by harvesting sustainable energy from the natural environment. By combining the charge leakage effect and the ternary dielectric triboelectrification effect, it improves the performance of direct-current TENGs. The optimized T-DC-TENG achieves an ultra-high average power density of 6.15 W m(-2) Hz(-1) and a historically low crest factor (1.0043), surpassing previous reports.

Summary: Using first-principles calculations, we predict that two ferromagnetic monolayers, BiXO3 (X = Ru, Os), have direct band gaps and high Curie temperatures. Monte Carlo simulations show that the monolayers exhibit high Tc beyond 400 K. Both monolayers demonstrate out-of-plane magnetic anisotropy, with significant magnetic anisotropy energies. The electronic and magnetic properties of BiXO3 monolayers make them promising candidates for nanoscale electronics and spintronics.
