Summary: An efficient method for the synthesis of [1,4]thiazino[4,3-alpha]indole derivatives was developed using sodium chlorodifluoroacetate and elemental sulfur as the difluoromethylthiolating reagent system. The sequential transformation process selectively formed four bonds including one C-N, two C-S, and one C-C bonds in good yields.

Summary: The study shows that inactive methyl carbons can act as nucleophiles, attacking cationic centers and aiding in the synthesis of cyclopropane rings in certain triterpene backbones. Unconventional chemical processes were used to synthesize compounds with bridgehead cyclopropane or an 11-membered bicyclic structure within a triterpene skeleton. The proposed alpha-fluorocarbocation mechanism was supported by density functional theory calculations.

Summary: This article focuses on the use of TiO2 as a light-absorbing semiconductor for photocatalytic solar energy conversion and various photocatalytic reactions. Different strategies are discussed to improve the photoresponse of TiO2-based materials, including bandgap engineering, surface sensitization, nanostructuring, and geometry optimization. The article provides a comprehensive overview of the effects of fluorine on TiO2, including morphology modifications, and the influence of surface or bulk fluorine on the photocatalytic performance of TiO2. The challenges and future prospects for the development of fluorine effects are highlighted.

Summary: A new N-F reagent, N,N'-difluoro-2,2';-bipyridinium bis(tetrafluoroborate), for iodoarene-catalyzed fluorination reactions as a terminal oxidant is reported. This reaction is effective for the fluorination of simple alkenes, aromatic alkenes and 1,3-dicarbonyl compounds, giving 1,2-difluorinated alkanes, geminal difluorinated compounds, 2-fluorinated-1,3-dicarbonyl compounds in good to high yields, respectively. The present N-F reagent can be recovered as 2,2'-bipyridine.

Summary: An innovative one-step synthesis method for beta-aryl-beta,beta-difluorocarbamates and beta-aryl-beta,beta-difluoroazides is presented. The method involves a radical addition/fluorination reaction between alpha-fluorostyrene substrates and N-centered radicals in the presence of Selectfluor (R). The azidofluorination reaction showed the best results, and the resulting beta-aryl-beta,beta-difluoroazides were further transformed into nitrogen-based heterocycles and more complex azide derivatives.

Summary: Phosphoranides are hypervalent species that can serve as model compounds for intermediates or transition states in nucleophilic substitution reactions. This study reports the syntheses and properties of stable trifluoromethylphosphoranide salts, and investigates their reactivity towards electrophiles. The structures and dynamics of these compounds are determined using X-ray crystallography and NMR spectroscopy.

Summary: The regioselective difluoromethane sulfonylation or triflylation of resorufin derivatives provides access to 2-difluoromethane sulfonylated or triflylated resorufin derivatives in good yields. The addition of a difluoromethane sulfonyl group enhances the solubility of the chromophore and expands its Stokes shift, allowing for improved imaging of enzyme activity in live cells using a difluoromethane sulfonylated resorufin-based fluorogenic probe.

Summary: Administration of the fluorinated drug cinacalcet in rats resulted in tissue-specific fluorination of proteins, with three fluorinated proteins identified in the liver and four in the brain tissue after 21 days of treatment. Summary: The direct trifluoromethylthiolation of aziridines with AgSCF3 and iodides is reported. The beta-trifluoromethylthiolated isothiocyanates and amines were selectively obtained by changing the cation of iodide. This strategy shows good tolerance to a wide range of functional groups with high yields and regioselectivities. Moreover, the obtained isothiocyanates can be used for further synthetic manipulations.

Summary: This article describes two routes for the synthesis of potent and selective bromodomain and extra-terminal BD2 inhibitors, cis or trans C2,3,5,7-tetrasubstituted dihydrobenzofurans, followed by the optimization of the synthesis of the lead molecule GSK973 to support pre-clinical efficacy and safety studies. The use of flow chemistry, extensive optimization of the fluorination step, and high-yielding aminocarbonylation were critical in generating the required material. The identified new route also serves as a strong starting point for further optimization.

Summary: Trifluoromethyl ynones derived from HFO-1234yf react with nucleophiles and form heterocyclic derivatives via intramolecular reactions. The reaction with 2-amino and 2-hydroxyl benzaldehyde or ketone substrates gives trifluoromethyl quinoline and chromene products, offering a simple and economical approach to pharmaceutically relevant structures. The unusual fluxional behavior of the 2-trifluoromethylquinoline products is studied through 2-D NMR and X-ray crystallography.
