Aberrant amplification of the crosstalk between canonical Wnt signaling and N-glycosylation gene DPAGT1 promotes oral cancer

Oral cancer is one of the most aggressive epithelial malignancies, whose incidence is on the rise. Previous studies have shown that in a subset of human oral squamous cell carcinoma (OSCC) tumor specimens, overexpression of the DPAGT1 gene, encoding the dolichol-P-dependent N-acetylglucoseamine-1-phosphate transferase, a key regulator of the metabolic pathway of protein N-glycosylation, drives tumor cell discohesion by inhibiting E-cadherin adhesive function. Recently, we reported that DPAGT1 was a target of the canonical Wnt signaling pathway. Here, we link overexpression of DPAGT1 in human OSCC tumor specimens to aberrant activation of canonical Wnt signaling. We report dramatic increases in beta- and gamma-catenins at the DPAGT1 promoter and correlate them with reduced expression of a Wnt inhibitor, Dickkopf-1 (Dkk-1). Using human squamous carcinoma cell lines of the head and neck, we show that partial inhibition of DPAGT1 reduces canonical Wnt signaling, indicating that DPAGT1 and canonical Wnt signaling function in a positive feedback loop. We provide evidence that E-cadherin inhibits DPAGT1, canonical Wnt signaling and the OSCC cancer phenotype by depleting nuclear beta- and gamma-catenins, with hypoglycosylated E-cadherin being the most effective. This suggests that in human OSCC, extensive N-glycosylation of E-cadherin compromises its ability to inhibit canonical Wnt signaling and DPAGT1 expression. Our studies reveal a novel interplay between DPAGT1/N-glycosylation and canonical Wnt signaling and suggest that dysregulation of this crosstalk is a key mechanism underlying OSCC. They also suggest that partial inhibition of DPAGT1 may represent an effective way to restore normal interactions among these essential pathways in oral cancer. Published by Elsevier Ltd.

Occurrence and fate of Adsorbable Organic Halogens (AOX) in two WWTPs from Romania

Summary: This study analyzed the presence, occurrence, and composition of absorbable organic halogens in wastewater and sewage sludge, as well as their genotoxicity effect on the environment. The results showed that the AOX parameter was correlated with the concentration level of volatile organic compounds, chlorophenols, organochlorine pesticides, and polychlorinated biphenyls. The identified halogenated organic compounds accounted for only a small percentage of the total AOX amount, except in dehydrated sludge samples where they accounted for a significant proportion.

Oxidative Stress and Histopathological Changes in Gills and Kidneys of Cyprinus carpio following Exposure to Benzethonium Chloride, a Cationic Surfactant

Summary: This study evaluated the acute toxicity of benzethonium chloride (BEC) on the gills and kidneys of European carp. The results showed that BEC induced toxicity in these organs, as evidenced by altered antioxidant enzyme activities, increased lipid peroxidation, and decreased glutathione concentration. Histological observations revealed structural changes, particularly in the gills where the most severe effects were observed.

Biodegradation of two organic ultraviolet-filters by single bacterial strains

Summary: The presence of organic ultraviolet filters, such as 4-hydroxybenzophenone (4-HBP) and 2,4-dihydroxybenzophenone (BP-1), in the environment poses a threat to aquatic organisms. This study investigated the biodegradation potential of these compounds by different bacterial strains and found that the degradation rates varied among strains. These findings provide important insights into the biodegradation pathways of organic ultraviolet filters.

Comprehensive clinical, genome and transcriptomic analysis of primary ghost cell odontogenic carcinoma

Summary: This study presents a comprehensive clinical, genomic, and transcriptomic analysis of primary ghost cell odontogenic carcinoma (GCOC). The results provide the first comprehensive molecular atlas for primary GCOC, including previously identified CTNNB1 mutation and novel alterations in MAP3K, EP300, and the 22q11.21 region. The transcriptome analysis reveals significant involvement of cytokine-cytokine receptor interaction and the PI3K-Akt signaling pathway. More GCOC cases should be compared to validate these findings for accurate clinical guidance.

Transoral robotic surgery in oropharyngeal squamous cell carcinoma: A comparative study between da Vinci Single-Port and da Vinci Xi systems

Summary: This study compares the outcomes of transoral resections using the da Vinci SP and da Vinci Xi systems for oropharyngeal squamous cell carcinoma (OPSCC). The results indicate that the da Vinci SP system has advantages in terms of reduced docking and console times, as well as improved rates of wide negative resection margins. However, there were no significant differences in complication rates or postoperative functional outcomes between the two systems.
