Background: Whey protein is a potential source of bioactive peptides. Based on findings from in vitro experiments indicating a novel whey derived peptide (NOP-47) increased endothelial nitric oxide synthesis, we tested its effects on vascular function in humans. Methods: A randomized, placebo-controlled, crossover study design was used. Healthy men (n = 10) and women (n = 10) (25 +/- 5 y, BMI = 24.3 +/- 2.3 kg/m(2)) participated in two vascular testing days each preceded by 2 wk of supplementation with a single dose of 5 g/day of a novel whey-derived peptide (NOP-47) or placebo. There was a 2 wk washout period between trials. After 2 wk of supplementation, vascular function in the forearm and circulating oxidative stress and inflammatory related biomarkers were measured serially for 2 h after ingestion of 5 g of NOP-47 or placebo. Macrovascular and microvascular function were assessed using brachial artery flow mediated dilation (FMD) and venous occlusion strain gauge plethysmography. Results: Baseline peak FMD was not different for Placebo (7.7%) and NOP-47 (7.8%). Placebo had no effect on FMD at 30, 60, and 90 min post-ingestion (7.5%, 7.2%, and 7.6%, respectively) whereas NOP-47 significantly improved FMD responses at these respective postprandial time points compared to baseline (8.9%, 9.9%, and 9.0%; P < 0.0001 for time x trial interaction). Baseline reactive hyperemia forearm blood flow was not different for placebo (27.2 +/- 7.2%/min) and NOP-47 (27.3 +/- 7.6%/min). Hyperemia blood flow measured 120 min post-ingestion (27.2 +/- 7.8%/min) was unaffected by placebo whereas NOP-47 significantly increased hyperemia compared to baseline (29.9 +/- 7.8%/min; P = 0.008 for time x trial interaction). Plasma myeloperoxidase was increased transiently by both NOP-47 and placebo, but there were no changes in markers inflammation. Plasma total nitrites/nitrates significantly decreased over the 2 hr post-ingestion period and were lower at 120 min after placebo (-25%) compared to NOP-47 (-18%). Conclusion: These findings indicate that supplementation with a novel whey-derived peptide in healthy individuals improves vascular function.

Vitamin A and D Absorption in Adults with Metabolic Syndrome versus Healthy Controls: A Pilot Study Utilizing Targeted and Untargeted LC-MS Lipidomics

Summary: Individuals with metabolic syndrome (MetS) absorb less vitamin E compared to healthy controls. This study found that MetS subjects have significantly higher absorption of vitamin A, while the absorption of vitamin D-2 is less affected by MetS status. The association between phospholipids and vitamin A suggests that enzymes may play a role in the higher bioavailability of vitamin A in MetS subjects.

Catechin-Rich Green Tea Extract and the Loss-of-TLR4 Signaling Differentially Alter the Hepatic Metabolome in Mice with Nonalcoholic Steatohepatitis

Geoffrey Y. Sasaki, Jinhui Li, Morgan J. Cichon, Rachel E. Kopec, Richard S. Bruno

Summary: The study shows that catechin-rich green tea extract (GTE) limits inflammation in nonalcoholic steatohepatitis (NASH) through a Toll-like receptor 4 (TLR4)-dependent mechanism. GTE supplementation during NASH shifts the hepatic metabolome similarly to the loss-of-TLR4 signaling. However, the effects of GTE on hepatic metabolism are distinct from those of loss-of-TLR4 signaling, suggesting different mechanisms for the anti-inflammatory activities of GTE and loss-of-TLR4 signaling in abrogating NASH.

Application of Comparative Lipidomics to Elucidate Postprandial Metabolic Excursions Following Dairy Milk Ingestion in Individuals with Prediabetes

Li Chen, Shiqi Zhang, Xiaowei Sun, Joshua D. McDonald, Richard S. Bruno, Jiangjiang Zhu

Summary: This study investigated the differential plasma lipidomic responses to non-fat or full-fat milk ingestion during postprandial hyperglycemia and found that dairy milk, especially non-fat milk, protected against lipid changes induced by glucose ingestion. Co-ingestion of dairy milk with glucose significantly altered lipid profiles, with full-fat milk more substantially modulating lipid profiles, suggesting dairy milk-mediated lipid modulation may be an effective dietary approach to reduce the risk of metabolic diseases among those with prediabetes.

Summary: The study demonstrates that liver inflammation combined with spinal cord injury exacerbates intraspinal pathologies, including non-alcoholic steatohepatitis, and impairs locomotor recovery. Liver-derived protein Fetuin-A may act as a potential neuropathological mediator.

Daily Inclusion of Resistant Starch-Containing Potatoes in a Dietary Guidelines for Americans Dietary Pattern Does Not Adversely Affect Cardiometabolic Risk or Intestinal Permeability in Adults with Metabolic Syndrome: A Randomized Controlled Trial

Sisi Cao, Emily L. Shaw, William R. Quarles, Geoffrey Y. Sasaki, Priyankar Dey, Joanna K. Hodges, Avinash Pokala, Min Zeng, Richard S. Bruno

Summary: Including potatoes, which are rich in resistant starch, in a dietary pattern based on the Dietary Guidelines for Americans (DGA) can improve cardiometabolic and gut health in individuals with metabolic syndrome. Short-term consumption of a DGA-based diet reduces cardiometabolic risk, and the addition of resistant starch-containing potatoes to a healthy diet reduces small intestinal permeability and postprandial endotoxemia.

Dairy Milk Casein and Whey Proteins Differentially Alter the Postprandial Lipidome in Persons with Prediabetes: A Comparative Lipidomics Study

Summary: This study found that whey protein has an earlier and stronger impact in maintaining the stability of the lipidome compared to casein protein. Whey can protect against glucose-induced changes in glycerophospholipid and sphingolipid metabolism, while ether lipid metabolism and SP metabolism are the pathways most greatly impacted by casein.

Summary: Vitamin C is an essential micronutrient that enhances antibody response by promoting plasma cell differentiation through IL-21/STAT3 signaling pathway. As a cofactor for epigenetic enzymes, it facilitates DNA modification and demethylation, thereby influencing gene expression and cell fate decisions in the immune system.

Catechin Bioavailability Following Consumption of a Green Tea Extract Confection Is Reduced in Obese Persons without Affecting Gut Microbial-Derived Valerolactones

Summary: Obesity decreases the bioavailability of catechins, but does not affect valerolactone generation and urinary elimination of catechins. The composition of gut microbiota is altered in obese individuals. The levels of ascorbic acid and gut microbiota diversity are correlated with the bioavailability of catechins.
