Summary: This study investigates the alternative splicing of tau protein in HD patients and identifies abnormal splicing events in the putamen. Differences in exon 2 splicing coincide with increased tau hyperphosphorylation, aggregation, and markers of neurodegeneration, suggesting a potential biomarker or therapeutic target.

Summary: Through research on AD patient samples and mouse models, it was found that there is a parallel association between PrPC and the 4R isoforms of tau protein. Reducing or eliminating PrPC levels leads to an increase in the balance of tau protein 3R/4R.

Summary: The study shows that phosphorylation of TDP-43 at specific sites by CK1 epsilon can impact its function in tau mRNA processing, with blocking mutations leading to increased cytoplasmic aggregation of TDP-43 and mimicking mutations enhancing this effect.

Summary: The study utilized a splicing-guided aggregation method to investigate the functional impact of multiple low-frequency TREM2 variants on alternative splicing in Alzheimer's disease, revealing an exon skipping event in the second exon of TREM2 in individuals with a low-frequency variant causing loss of function for a splicing regulatory element. Genes differentially expressed between AD patients with high vs. low skipping of the second exon were significantly enriched in immune-related pathways. This method provides new insights into the regulation of alternative splicing of TREM2 by low-frequency variants and could aid in exploring potential molecular mechanisms of disease-related variants.

Summary: This study found that hnRNPA1 affects the migration, invasion, and EMT transition of lung cancer cells by regulating the splicing of LAS1L gene. The expression of hnRNPA1 in lung cancer tissues showed heterogeneity and may provide new directions for research and potential therapeutic targets in lung cancer metastasis.

Summary: Steroid hormones not only regulate gene transcription but also modulate the alternative splicing of pre-mRNA, acting through binding to nuclear receptors. The effect of steroid hormones on alternative splicing is time-, gene-, and tissue-specific. Mechanisms by which steroid hormones regulate alternative splicing include the recruitment of dual-function proteins, transcriptional regulation of splicing factor levels, alternative splicing of splicing factors or transcription factors, and regulation of elongation rate.

Summary: NF-YA, the regulatory subunit of the trimeric CCAAT-binding transcription factor NF-Y, has multiple alternative spliced isoforms, and NF-YAx and NF-YAg are enriched in Aves and play a major role in early chicken embryo development. In addition, the absence of exon-5 in NF-YAg leads to a global reduction of beta-stranded elements, while removal of the disordered exon-3 sequence has limited effects on TAD architecture.

Summary: Understanding the regulation of MAPT splicing is crucial for unraveling the pathogenesis of tauopathies, such as AD and PSP, where different tau isoforms accumulate. The study revealed the importance of N-terminal splicing and its coordination with exon 10 inclusion/exclusion in tauopathies.

Summary: Alternative splicing in Tau exon 10 generates 3 R- and 4 R-Tau proteoforms, which are equally abundant in healthy adult human brain. Aberrant splicing in Tau exon 10 can lead to imbalanced expression levels of 3 R- and 4 R-Tau, triggering toxic Tau aggregation, neuron dysfunction, and patient death in tauopathies. This study identifies hnRNPC as a regulator that promotes Tau exon 10 inclusion by interacting with U-tract motifs in introns 9 and 10 of Tau pre-mRNA, suggesting a potential therapeutic role for hnRNPC in progressive supranuclear palsy (PSP), a tauopathy.

Summary: Alternative splicing is a highly combinatorial mechanism that allows higher eukaryotes to increase mRNA diversity. The selection of alternative splice sites is influenced by sequence, cis-regulatory elements, exon and intron length, and RNA secondary structures. The study showed that the mode of splice site recognition, either intron or exon definition, plays a role in alternative splicing decisions.
