The nuclear factor-kappa B (NF-kappa B) family of regulators is well known for mediating dendritic cell (DC) maturation-that is, the acquisition of the functions required for full activation of T cells. Paradoxically, a key member of this family, NF-kappa B1, is now also implicated in maintaining DCs in an immature state (pages 1663-1667).

Epitope-based precision immunotherapy of Type 1 diabetes

Summary: Antigen-specific immunotherapies (ASITs) are important in treating autoimmune diseases, but their effectiveness is influenced by patient characteristics in Type 1 diabetes. Precision medicine targeting specific T cell populations and patient subgroups is a new goal. Profiling methods for antigen-specific T cells need improvement, as well as delivery approaches for tolerance induction. The field is moving toward using patient-specific variations to improve efficacy, but challenges remain.

Summary: The study demonstrates the potential of using the intestine as a source for cellular reprogramming to replace lost pancreatic beta cells in diabetes. By ablating FoxO1, the Paneth/goblet cell lineage can be converted into insulin-producing cells. Through a combination treatment targeting FOXO1, Notch, and TGF-beta, near normalization of glucose levels can be achieved in diabetic animals, accompanied by the generation of intestinal insulin-producing cells.
