Engineering ergothioneine production in Yarrowia lipolytica

Untargeted metabolomics of COVID-19 patient serum reveals potential prognostic markers of both severity and outcome

Summary: This study utilizes untargeted metabolomics analysis to evaluate 120 samples of COVID-19 patients and predict the severity and outcome of the infection. The study identifies a group of metabolites with relevant biological functions, such as deoxycytidine and ureidopropionate (indirectly reflecting viral load), kynurenine (reflecting host inflammatory response), and multiple short chain acylcarnitines (energy metabolism) that can predict disease severity and outcome.

Summary: Fibrin amyloid microclots and platelet hyperactivation in South African patients with Long COVID/PASC may be responsible for the persistent symptoms. Hypertension, high cholesterol levels, cardiovascular disease, and type 2 diabetes mellitus were found to be the most important comorbidities. The study highlights the need for further research on the pathophysiology and treatment of Long COVID/PASC.

Membrane transporter identification and modulation via adaptive laboratory evolution

Summary: Membrane transport proteins are potential targets for medical and biotechnological applications. In this study, adaptive laboratory evolution was used to identify membrane transporters for four amino acids and specific mutations that modulate the activities of these transporters. The evolved strains displayed increased growth rates at high concentrations of the targeted amino acids. Whole genome sequencing revealed key mutations targeting the transporters and reverse engineering confirmed their causality. The functional roles of novel transporters were validated, and a production strain showed higher productivity when an identified exporter was overexpressed.

Alzheimer's Disease: A Systems View Provides a Unifying Explanation of Its Development

Summary: Alzheimer's disease (AD) is a neurodegenerative disorder with no cure, affecting 50 million people worldwide. The disease is characterized by the presence of senile plaques and neurofibrillary tangles, made up of amyloid-beta and hyperphosphorylated tau proteins. Despite various hypotheses, such as the amyloid-beta, inflammatory, vascular, and infectious hypotheses, there is still an incomplete understanding of AD pathogenesis. This paper presents a systems biology approach to connect these hypotheses and suggests P. gingivalis as a potential treatment target.

Relationship between the concentration of ergothioneine in plasma and the likelihood of developing pre-eclampsia

Summary: In this study, the concentration of the antioxidant nutraceutical ergothioneine in the plasma of 432 first time mothers was analyzed. Results showed a weak association between ergothioneine levels and maternal age, but not with BMI. Out of these 432 women, 97 developed pre-term or term pre-eclampsia. A threshold at the 90th percentile of the reference range in the control population was set, and it was found that only 1% of the women with ergothioneine levels above this threshold developed pre-eclampsia, suggesting a potential protective effect.

Increased Levels of Inflammatory and Endothelial Biomarkers in Blood of Long COVID Patients Point to Thrombotic Endothelialitis

Summary: The prevailing hypotheses for the persistent symptoms of Long COVID have been narrowed down to immune dysregulation and autoantibodies, widespread organ damage, viral persistence, and fibrinaloid microclots (entrapping numerous inflammatory molecules) together with platelet hyperactivation. We demonstrate significantly increased concentrations of von Willebrand factor (VWF), platelet factor 4 (PF4), serum amyloid A (SAA), a-2 antiplasmin (a-2AP), endothelial-leukocyte adhesion molecule 1 (E-selectin), and platelet endothelial cell adhesion molecule (PECAM-1) in the soluble part of the blood. Presence of microclotting, together with relatively high levels of six biomarkers known to be key drivers of endothelial and clotting pathology, points to thrombotic endothelialitis as a key pathological process in Long COVID.

Long COVID: pathophysiological factors and abnormalities of coagulation

Summary: Approximately 10% of COVID-19 patients experience prolonged symptoms known as Long COVID, affecting around 65 million people worldwide. Multiple pathophysiological processes contribute to Long COVID, including viral factors, host factors, and downstream impacts, all leading to the persistence of the disorder. These mechanisms result in the development of thrombotic endothelialitis, endothelial inflammation, hyperactivated platelets, and fibrinoid microclots, affecting all organ systems and explaining the various symptoms of Long COVID.

Are fibrinaloid microclots a cause of autoimmunity in Long Covid and other post-infection diseases?

Summary: It is now well established that the polymerisation of blood-clotting protein fibrinogen can lead to the formation of amyloid fibrin clots, known as fibrinaloids. These clots entrap molecules and can block microcapillaries, causing various diseases including Long COVID. The formation of fibrinaloids also generates novel epitopes, leading to the generation of autoantibodies and contributing to autoimmunity.

A Perspective on How Fibrinaloid Microclots and Platelet Pathology May be Applied in Clinical Investigations

Summary: Microscopy imaging can be used to investigate the properties of microclots and contribute to the clinical diagnoses and prognoses of different diseases. Measuring parameters such as size, distribution, spectral properties, resistance to proteolysis, cross-seeding ability, and concentration dependence of binding to small molecules can provide valuable information. Combining microscopy imaging with other methodologies like proteomics and imaging flow cytometry could open up new possibilities for personalized medicine.

Data sharing: A Long COVID perspective, challenges, and road map for the future

Summary: 'Long COVID' refers to the continued experience of SARS-CoV-2 symptoms in patients who have survived a COVID-19 infection. Millions of people worldwide are affected by Long COVID. Solving this conundrum requires lessons learned from the COVID-19 pandemic, where experts from various disciplines collaborated and shared data to tackle the problem. However, there is currently no global consensus on the definition, diagnosis, and treatment of Long COVID. This study examines data sharing and data science applications in understanding Long COVID and expediting relief for those affected.
