Seven pentacyclic triterpenoids including 3-O-(E)-coumaroyl-15-hydroxy--amyrin (1), 15-hydroxy--amyrin (2), 3-taraxerol (3), 3-taraxerol formate (4), 3-taraxerol acetate (5), 3-O-(E)-coumaroyl-taraxerol (6), and 3-O-(Z)-coumaroyl-taraxerol (7) were isolated and identified from the stems and twigs of the mangrove plant Rhizophora stylosa. The structures of the isolated compounds were determined by extensive analysis of their spectroscopic data. Among these metabolites, compound 1 is a new oleanane-type triterpenoid coumaroyl ester, while compound 4 is a new natural product obtained here as an isolated substance for the first time. The full spectroscopic detail of compound 4 is also reported here for the first time.

Mining new meroterpenoids from the marine red alga-derived endophytic Penicillium chermesinum EN-480 by comparative transcriptome analysis

Summary: The transcriptome analysis of the algal endophytic fungus Penicillium chermesinum EN-480 revealed that fermentation in modified rice solid medium could lead to the production of new meroterpenoids with different structures compared to other media. These compounds showed antimicrobial activity against human- and aqua-bacteria as well as plant-pathogenic fungi. The study also discussed the possible biosynthesis pathway of these compounds.

Cyclopiumolides A and B, unusual 13-membered macrolides from the deep sea-sourced fungus Penicillium cyclopium SD-413 with antiproliferative activities

Summary: This article reports the discovery of two novel biosynthetic related compounds, Cyclopiumolides A (1) and B (2), from the fungus Penicillium cyclopium SD-413 obtained from deep-sea sediments in the East China Sea. The structures of Cyclopiumolides A (1) and B (2) were determined through extensive NMR spectroscopic and mass spectrometric data analysis. Their relative and absolute configurations were determined using quantum mechanical calculations and NMR data calculations. Compounds 1 and 2 exhibited significant cytotoxic effects against tumor cell lines and molecular docking simulations were conducted to explore their inhibition modes and binding sites.

Aromatic Polyketides from the Deep-Sea Cold-Seep Mussel Associated Endozoic Fungus Talaromyces minioluteus CS-138

Summary: Five new aromatic polyketides, including a unique benzofuran derivative, talarominine A (1), and four chromone analogs talamins A-D (2-5), were isolated and identified from the endozoic fungus Talaromyces minioluteus CS-138 obtained from the deep-sea cold seep mussel Gigantidas platifrons. The chemical structures of these compounds were elucidated and their antibacterial and DPPH scavenging activities were evaluated, showing promising bioactivities. This work represents the first report on marine natural products of mussel-derived fungus living in cold seep environments.

Emestrin-type thiodiketopiperazines from Aspergillus nidulans SD-531, a fungus obtained from the deep-sea sediment of cold seep in the South China Sea

Summary: Three new emestrin-type thiodiketopiperazines and three known analogues were isolated and identified from a fungus obtained from deep-sea cold seep sediment. Their structures and absolute configurations were determined by spectroscopic analysis and TDDFT-ECD calculations. Compounds 3-6 showed antimicrobial activity, while compounds 3-5 exhibited cytotoxic activity against tumor cells. Compound 4 demonstrated the strongest cytotoxic activity with an IC50 value of 0.25 μM, surpassing the positive control sorafenib.

Rubensteroid A, a new steroid with antibacterial activity from Penicillium rubens AS-130

Summary: A new steroid, rubensteroid A (1), with strong antibacterial activity, and its decarboxylic analogue, solitumergosterol A (2), were isolated and identified from the Magellan Seamount-derived fungus Penicillium rubens AS-130. The structure and absolute configuration of compound 1 were determined by detailed NMR spectroscopic analysis, mass spectrometry data, and TDDFT-ECD calculations. Rubensteroid A (1) showed potent antibacterial activity against Escherichia coli and Vibrio parahaemolyticus, with MIC values of 0.5 μg/mL.

Antibacterial polyketides and ascochlorins from deep-sea cold-seep-derived fungus Furcasterigmium furcatum (syn. Acremonium furcatum)

Summary: Three new secondary metabolites, including a polyketide, an ascochlorin analogue, and a pair of enantiomers, along with nine known ascochlorins, were identified from a fungus isolated from deep-sea sediment samples. The structures were determined through NMR and mass spectrometric data interpretation. Compound 1 demonstrated potent activity against the human pathogenic bacterium Pseudomonas aeruginosa. The isolated ascochlorins also showed potent activities against aquatic bacteria, particularly Vibrio harveyi and V. alginolyticus.

Antimicrobial polyketides from Magellan Seamount-derived fungus Talaromyces scorteus AS-242

Summary: Seven new highly oxygenated natural products, including glucosidic polyketides, aromatic polyketides, azaphilone polyketides, and a drimane sesquiterpene lactone, were isolated from a fungus obtained from deep-sea cold-seep sediments. Some compounds were enhanced minor components and compounds 3 and 4 might be produced by the activation of silent gene clusters triggered by SAHA. Compound 7 exhibited potent activities against agricultural pathogenic fungi. This study provides a useful strategy for activating cryptic fungal metabolites from deep-sea-derived fungi.
