Aim: This study aimed to evaluate, for the first time the potential use of a safe biocompatible nanoformulation of silymarin (SM) as antitumor agent and to provide its mechanism of action compared with native SM. Materials & methods: SM was loaded into pluronic nanomicelles and Ehrlich ascites carcinomatumor-bearing mice were used as experimental model. Biochemical parameters including SOD, CAT and GSH, lipid peroxidation biomarkers (MDA), histopathological, ultrastructural and immunohistochemical studies were applied on the Ehrlich ascites carcinoma cells. Furthermore, the cell cycle as well as caspase-3 were examined. Results & conclusion: Nanoformulated SM (SMnp) destroyed tumors via increasing SOD, CAT and GSH concomitant with decreasing MDA. Moreover, SMnp-induced apoptosis through decreasing Ki-67 and Bcl2 expression, along with the activation of caspase-3, leads to inhibition of proliferation and the arrest of ceel cycle progression at the G1/S phase. Electron microscopy studies presented the superiority of SMnp over native SM in causing mitochondrial and nuclear degeneration in cancer cells.

Novel doxorubicin/folate-targeted trans-ferulic acid-loaded PLGA nanoparticles combination: In-vivo superiority over standard chemotherapeutic regimen for breast cancer treatment

Sahar A. Helmy, Saif El-Mofty, Amal M. El Gayar, Ibrahim M. El-Sherbiny, Yousra M. El-Far

Summary: The study aimed to enhance the efficacy of trans-ferulic acid (TFA) by loading it into folate-receptor-targeted poly lactic-co-glycolic acid nanoparticles (FA-PLGA-TFA NPs) and evaluating the antitumor efficacy of Doxorubicin (Dox)/FA-PLGA-TFA NPs combination against DMBA-induced breast cancer, as well as its safety profile. The combination not only inhibited Notch signaling but also suppressed synergy with other pathways, reduced P-gp levels, and maintained organ health and white blood cell count. This approach could potentially reduce side effects and improve antitumor activity compared to the standard AC regimen.

Drag-minimizing spore/pollen-mimicking microparticles for enhanced pulmonary drug delivery: CFD and experimental studies

Alaa M. Ali, Ahmed S. Abo Dena, Magdi H. Yacoub, Ibrahim M. El-Sherbiny

Summary: The aim of this work is to build a computational model to study the flowability of fungal spores and pollen grains in order to predict their ability to reach the deep lung and relate it to allergies. The results show that the drag force exerted on the spores and pollen grains can affect their ability to cause disease. Additionally, certain shapes of spores and pollen grains have low drag and can be used for drug delivery to the deep lung.

Voltammetric determination of Salmonella typhimurium in minced beef meat using a chip-based imprinted sensor

Shaimaa A. Khalid, Rabeay Y. A. Hassan, Rasha Mohamed El Nashar, Ibrahim M. El-Sherbiny

Summary: In this study, a bacterial imprinted polymer-based biosensor was developed for rapid and selective detection of Salmonella typhimurium. The biosensor exhibited high sensitivity and specificity towards the target pathogen and showed no interference signals when tested with other non-targeted pathogens. The biosensor also demonstrated the capability to directly detect the pathogen in minced beef meat samples, eliminating the need for bacterial enrichment steps.

Novel highly effective combination of naturally-derived quercetin and ascorbyl palmitate and their nanoformulations as an advancement therapy of cancer

Mohamed El -Far, Alaa Essam, Fardous F. El-Senduny, Amira O. Abd El-Azim, Sarah Yahia, Ibrahim M. El-Sherbiny

Summary: In this study, the anticancer effect of a novel drug combination using nanoformulations of Quercetin and Ascorbyl palmitate was investigated in Ehrlich ascites carcinoma (EAC)-bearing mice. The combination treatment significantly reduced tumor growth, increased antioxidant status, and reduced lipid peroxidation. The combination also downregulated the expression of p-STAT3 and HIF-1 alpha, induced cell cycle arrest and apoptosis in EAC cells, and caused DNA damage. The combination was also superior in the destruction of the mitochondrial membrane.

A comprehensive review on lipid nanocarrier systems for cancer treatment: fabrication, future prospects and clinical trials

Mohamed Fawzi Kabil, Osama A. Badary, Frank Bier, Shaker A. Mousa, Ibrahim M. El-Sherbiny

Summary: Cancer is a major cause of death worldwide, and chemotherapy is the most commonly used treatment. However, current chemotherapeutic treatments have limitations, including lack of specificity and adverse effects. Lipid nanoparticles (LNPs) offer a promising nanocarrier system to improve drug delivery and reduce side effects, potentially leading to better therapeutic outcomes.
