Summary: Due to the lack of pandemic potential, research on the pathology and immunological responses of IBV lags behind that of IAV. The impact of lifestyle and environmental factors, such as cigarette smoking, on IBV infections is still unclear. By pre-treating mice with cigarette smoke extract for two weeks, infecting them with IBV, and monitoring the effects, we developed an animal model system. The results showed that cigarette smoke extract treatment decreased IBV specific IgG levels but did not change viral replication and weight recovery post infection in the upper airway/lung. However, higher concentrations of cigarette smoke extract did lead to higher mortality post infection, suggesting that inflammation induced by smoking coupled with IBV infection resulted in exacerbated disease outcome.

Summary: Natural killer (NK) cells, important in early defense against tumors and virally infected cells, are regulated by a balance of activating and inhibitory receptors. However, some activating receptors lack an inhibitory partner. This study shows that activating NK cell receptors without inhibitory partners are not a "master" signal, but instead their activity is controlled through regulation of other activating receptors. This finding has potential implications for the development of NK cell-based cancer immunotherapy.

Summary: The study reveals that human colonic ILC3s can express granzyme B in response to a subset of enteric bacteria through a process mediated by IL-15. This finding raises new questions about the multifunctional role of human gut ILC3s.

Summary: Cigarette smoke can damage a wide range of immunological functions, including innate and adaptive immune responses. The inflammasome complex is crucial in innate immune response, particularly in cigarette smoke-related diseases and physiopathological disorders. Strategies targeting the ROS/NLRP3 inflammasome axis show potential therapeutic effects in these contexts.

Summary: Only three well-characterised cases of functional cure have been described in paediatric HIV infection over the past decade. This underlines the fact that early initiation of combination antiretroviral therapy (cART), whilst minimising the size of the viral reservoir, is insufficient to achieve cure, unless other factors contribute. HIV-specific cytotoxic T-lymphocyte (CTL) and natural killer (NK) cell responses play crucial roles in functional cure in paediatric infection.

Summary: The liver plays a crucial role in systemic immunity and defends against the spread of tumor cells. It contains a higher percentage of innate immune lymphocytes, such as NK and NKT cells, which eliminate malignant cells with their strong cytotoxic ability. These lymphocytes also initiate immune responses by adaptive immune cells, making them important contributors to systemic antitumor immunity. Understanding the mechanisms of these immune cells provides insight for tumor immunity research and potential clinical applications for cancer therapies.

Summary: Immunization with Ft LPS triggers production of IL-25, which promotes activation and differentiation of B1 cells into IgM secreting cells through stimulation of IL-5 release by ILC2. This IL-25-ILC2-IL-5 axis represents a potential novel strategy to improve vaccination against T-independent bacterial antigens.

Summary: NK cells are known to have cytotoxic effects on tumor cell lines, but their specific roles in primary tumor detection and elimination remain unclear. ILC1 play an active role in inhibiting the antitumoral immune response, suggesting the need to evaluate the tumor infiltration of ILC1 and NK cells to optimize immune harnessing in cancer therapies.

Summary: Interleukin-1 (IL-1) is a cytokine that plays a significant role in inflammation and immune response, primarily produced by macrophages during defensive reactions. It belongs to a superfamily of structurally similar proteins and acts through binding to specific receptors on the plasma membrane of target cells. IL-1 family cytokines not only have innate immune and inflammatory effects but also appear to be involved in the development of adaptive immunity in vertebrates.

Summary: In this study, we evaluated the efficacy and immune effects of cetuximab plus avelumab combination treatment in NSCLC patients who had previously responded to PD-1 inhibitors. The results showed that this combination treatment can induce anti-tumor activity and activate the ADCC mediated by NK cells as well as the STING pathway.

Summary: Chronic inflammation has been identified as a risk factor for human cancer, and this study showed that cigarette smoke-induced genomic instability may be partially attributed to IL-17-mediated inflammation. Higher levels of IL-17 and DDR were observed in smokers' lung tissues compared to non-smokers, as well as in mice exposed to cigarette smoke. These findings suggest that IL-17 could play a crucial role in the development of lung cancer.
