Summary: USP11 is significantly upregulated in hepatocellular carcinoma (HCC) and is closely associated with tumor number, vascular invasion, and poor prognosis. USP11 promotes HCC metastasis and epithelial-mesenchymal transition (EMT) by inducing the transcription factor Snail. Mechanistically, USP11 interacts with and deubiquitinates eEF1A1, leading to increased expression of HGF and activation of the PI3K/AKT signaling pathway through the eEF1A1/SP1/HGF axis. Thus, USP11 could be a potential therapeutic target for the treatment of HCC.

Summary: Zinc finger protein 275 (ZNF275) has been found to be highly expressed in cervical cancer tissues and downregulation of ZNF275 can inhibit tumor growth and weaken the AKT/Bcl-2 signaling pathway. Additionally, the AKT inhibitor triciribine synergizes with cisplatin to enhance the therapeutic effect on cervical cancer cells expressing high ZNF275. The combination treatment of triciribine and cisplatin is more effective in inducing tumor regression than single agents in cervical cancer PDX models expressing high ZNF275.

Summary: Cisplatin is commonly used for cancer treatment but can cause irreversible sensorineural hearing loss. A new compound called aucubin has been found to attenuate cisplatin-induced damage to cochlear hair cells through various mechanisms, including reducing apoptosis, reactive oxygen species production, and mitochondrial dysfunction. In vivo experiments also showed that aucubin protected against cisplatin-induced hearing loss and hair cell loss. The protective effects of aucubin were found to be mediated by the STAT3 signaling pathway via PI3K/AKT activation.

Summary: ATG4D, a member of the ATG4 family, is upregulated in HCC and its silencing can suppress cell proliferation, promote apoptosis, and enhance sensitivity to cisplatin in MHCC-97L cells. Silencing of ATG4D also decreases Akt phosphorylation and increases caspase-3 protein level. These findings suggest that ATG4D may serve as a therapeutic target for HCC therapy.

Summary: Chemotherapy induces tumor cell death and inhibits tumor progression, but the accompanying immune responses in the surrounding dying tissue cause significant inflammation. This study found that cotreatment of 1-palmitoyl-2-linoleoyl-3-acetyl-rac-glycerol (PLAG) with the AC regimen chemotherapy enhanced the antitumor effect in a mouse model of triple-negative breast cancer. PLAG inhibited excessive neutrophil infiltration and alleviated side effects of chemotherapy. Reduced neutrophils and increased lymphocytes in PLAG-treated mice maximized the antitumor effect and inhibited tumor growth.
