Summary: The metformin-decavanadate complex inhibits Ca2+-ATPase through a mixed-type competitive-non-competitive inhibition, with a higher IC50 value compared to decavanadate. Both decavanadate and metformin-decavanadate exhibit antiproliferative effects on melanoma cells at lower concentrations than monomeric vanadate. The compounds also increase phosphorylation of ERK and AKT signaling proteins, suggesting the anti-proliferative activities are independent of growth-factor signaling pathways.

Summary: Small nucleotide variants in non-coding regions of the genome can activate oncogenic gene regulatory networks by altering transcriptional regulation. Melanoma has a high burden of non-coding variants, and a pipeline was developed to identify hotspot regions containing putatively functional non-coding somatic variants. Through the study of a hotspot in the promoter of CDC20, it was found that variants disrupting an ETS motif led to downregulation of CDC20 and perturbation of key melanoma phenotypes.

Summary: Erucin (ERU), a major isothiocyanate in rocket salads, has shown high efficacy as an anticancer agent. Recent studies have revealed that ERU has chemopreventive effects and acts as a H2S donor in human pancreatic adenocarcinoma. This study demonstrated that ERU can inhibit melanoma cell proliferation, induce apoptosis and cell cycle arrest, and inhibit migration, invasiveness, and clonogenic abilities. These effects are associated with melanogenesis impairment and mitochondrial fitness modulation.
